摘  要:目的:探讨二乙酰二脱水卫矛醇(diacetyldianhydrogalactitol,DADAG)对小鼠白血病L1210细胞凋亡相关蛋白表达的影响。方法:MTT法测定DADAG对L1210细胞的杀伤作用;流式细胞技术和透射电镜检测细胞凋亡;Western blot法分析细胞内Bcl-2和Bcl-X_L蛋白水平的变化。结果:与对照组相比,DADAG可降低L1210细胞的存活率,且具有浓度依赖性。给药12.0、17.2、24.5、35.0和50.0mg/L 24h后,流式细胞术周期分析结果显示,各处理组均出现凋亡峰。DADAG 50.0mg/L作用L1210细胞24h后,靶细胞出现固缩、染色质边集;DADAG可下调Bcl-2和Bcl-X_L蛋白水平,且具有时间依赖性。结论:DADAG可能通过抑制Bcl-2及Bcl-X_L蛋白的表达而诱导L1210细胞发生凋亡。Objective: To investigate the effect of diacetyldianhydrogalactitol (DADAG) on expression of apoptosis-related protein Bcl-2 and Bcl-XL. in mouse leukemia L1210 cells. Methods:The cytotoxic effect of DADAG on L1210 cells was determined using MTT assay. DADAG-induced apoptosis in L1210 cells was identified by flow cytometry and electron microscopy. The levels of Bcl-2 and Bcl-XL, proteins were examined by western blot analysis, Results: DADAG dose-dependently decreased the survival rates of L1210 cells compared with control. Apoptotic peaks were detected by flow cytometry after 24-h treatment with DADAG at the concentrations of 12.0, 17.2, 24.5, 35.0, and 50.0 mg/L. Treatment with DADAG 50.0 mg/L for 24 h induced shrinkage of cytoplasm and condensation and margination of chromatin. DADAG down-regulated the expression of Bcl 2 and BclXL proteins in a time-dependent manner. Conclusion: DADAG induces apoptosis of L1210 cells via inhibiting the expressions of Bcl-2 and Bcl-XL proteins.
