摘  要:目的研究利塞膦酸钠对人成骨样细胞MG63护骨素(OPG)及肿瘤坏死因子α(TNFα)表达的影响,以阐明利塞膦酸钠发挥骨保护作用的机制。方法以不同浓度(10-10molL;10-9molL;10-8molL;10-7molL)的利塞膦酸钠干预MG63细胞72h;ELISA法测定细胞条件培养液中OPG及TNFα的含量。结果利塞膦酸钠可下调MG63细胞TNFα的表达;上调OPG的表达;并提高碱性磷酸酶(ALP)活性。结论利塞膦酸钠可通过调节成骨样细胞MG63OPG、TNFα的表达,从而发挥其抗骨吸收的作用。Objective To examine the effect of Risedronate on the expression of tumor necrosis factor alpha (TNFα) and osteoprotegrin (OPG) in human osteoblastic-like cell lines MG 63.Methods MG 63 cell lines were cultured with different concentrations of Risedronate (10^-10 mol/L; 10^-9 mol/L; 10^-8 mol/L; 10^-7 mol/L) for 72 h and enzyme-linked immunosorbent assay was performed to detect the changes of the expression of TNFα and OPG in conditioned media. Results OPG in conditioned media was dose-dependently upregulated (9753.0 ± 790. 1 vs 5618.5 ± 472. 1 pg/mg·pro) by Risedronate, while TNFα was dose-dependently down-regulated (3. 1 ± 0.8 vs 7. 1 ± 1.0 pg/mg · pro). ALP activity was also increased after Risedronate pretreatment. Conclusions Risedronate may inhibit osteoclastogenesis through up-regulating the expression of OPG, and down-regulating the expression of TNFα in human osteoblast-like cell lines MG 63.
