摘  要:目的:比较不同浓度的咪唑斯汀对体外培养皮肤组织中炎症因子表达的影响,以验证其对抗变态反应性炎症的作用。方法:实验于2003-10/2004-05在第三军医大学西南医院皮肤科实验室完成。将0,50,100和150μL的咪唑斯汀和皮肤组织体外共同培养,同时加入组胺、花生四烯酸等致炎刺激物,培养24h后反转录聚合酶链反应法测定上述药物作用后皮肤组织中单核细胞趋化蛋白1、单核细胞趋化蛋白3、活化后可调节的正常T细胞表达分泌的因子、嗜酸性粒细胞趋化因子等趋化因子mRNA表达,酶联免疫吸附试验法测定培养上清液中细胞因子白细胞介素4、白细胞介素5和肿瘤坏死因子α等炎症介质的含量,观察不同药物对其影响的情况。结果:①各组间的目的基因单核细胞趋化蛋白3/β-肌动蛋白、单核细胞趋化蛋白1/β-肌动蛋白、嗜酸性粒细胞趋化因子/β-肌动蛋白及活化后可调节的正常T细胞表达分泌的因子/β-肌动蛋白的检测值相差显著(咪唑斯汀0,50,100和150μL组目的基因与内参灰度比值分别为1.370±0.0025,1.301±0.0070,0.205±0.0016,0.555±0.0008;1.043±0.0035,1.005±0.0080,0.155±0.0020,0.393±0.0010;0.680±0.0020,0.705±0.0040,0.105±0.0010,0.285±0.0003;0.350±0.0030,0.525±0.0003,0.060±0.0020,0.215±0.0003,前三个指标P<0.01)。②培养上清液中各组间白细胞介素4、白细胞介素5和肿瘤坏死因子α三种细胞因子的测定值相差非常显著[咪唑斯汀0,50,100和150μL组分别为(302.105±7.085),(395.645±10.124),(205.879±12.354)μg/L;(251.124±10.520),(324.124±9.755),(142.347±9.017)μg/L;(198.645±7.332),(257.386±12.586),(112.540±7.846)μg/L;(135.683±6.531),(185.237±5.752),(89.232±11.579)μg/L,P<0.01]。结论:咪唑斯汀能明显抑制各种炎症因子的表达,并且浓度越高抑制效应越强,有明显的剂量依赖关系。AIM: To compare the effects of different concentration mizolastine on the expression of inflammatory factors of cultivated skin in vitro so as to verify the effect on allergic reaction inflammation. METHODS: The experiment was conducted at the Laboratory of Department of Dermatology,Southwest Hospital of Third Military Medical University of Chinese PLA from October 2003 to May 2004. The skin was co-cultivated with 0,50,100 and 150 μL mizolastine, and then histamine and arachidonic acid were added. Twenty-four hours later the mRNA expression levels of monocyte chemotactic protein-1 (MCP-1), monocyte chemotactic protein-3 (MCP-3), regulated on activation, normal T cell expressed and secreted (RANTES) and eotaxin were analyzed with reverse transcription-polymerase chain reaction (RT-PCR). The concentrations of interlukin (IL)-4, IL-5 and tumor necrosis factor-α (TNF-α) in cultivated superuatant were detected by enzyme kinked immunosorbent (ELISA) methods. Influences of different drugs were observed. RESULTS: ①There were significant difference in MCP-3/β-actin, MCP- 1/β-actin, eotaxin/β-actin and adjustable RANTES/β-actin (in the 0,50, 100 and 150 μL mizolastin groups ratio of objective gene to internal reference gray were 1.370±0.002 5,1.301 ±0.007 0,0.205±0.001 6,0.555 ±0.000 8 ; 1.043±0.003 5,1.005±0.008 0,0.155±0.002 0,0.393±0.001 0; 0.680 ±0.002 0,0.705 ±0.004 0,0.105 ±0.001 0,0.285 ±0.000 3;0.350 ±0.003 0,0.525±0.000 3,0.060±0.0020,0.215±0.000 3, respectively the former three indexes .P 〈 0.01). ②There were significantly differences in IL-4, IL-5 and TNF-α in cultured superuatant [in the 0,50,100 and 150 μL mizolastin groups it was (302.105±7.085), (395.645±10.124), (205.879 ±12.354) μg/L;(251.124±10.520),(324.124±9.755),(142.347±9.017) μg/L; ( 198.645±7.332), (257.386±12.586), (112.540±7.846)μg/L; ( β5.683 ±6.531 ), (185.237±5.752), (89.232±11.579) μg/L ,P 〈 0.01 ].
