摘 要:目的探讨高压氧对庆大霉素耳毒性的影响。方法36只豚鼠随机分为4组,每组9只18耳。对照组腹腔注射生理盐水,庆大霉素组腹腔注射庆大霉素150 mg/(kg·d),连续注射5 d。高气压组和高压氧组分别于每天注射庆大霉素之前行高气压和高压氧预暴露(0.2 MPa)1次,连续5 d。于实验前1 d和实验第6天分别以听觉脑干诱发电位(ABR)检测耳蜗听功能,并且于第6天检测后处死动物,行耳蜗毛细胞扫描电镜观察。结果高压氧预暴露组ABR阈移与生理盐水对照组差异不显著(P>0.05),与高气压组及庆大霉素组比较差异显著(P<0.01),耳蜗毛细胞改变与ABR检测结果一致。结论高压氧预暴露可以在近期内有效地保护豚鼠耳避免受庆大霉素耳毒性的损害。Objective To explore the effect of hyperbaric oxygen on gentamicin-induced ototoxicity in guinea pigs. Methods Thirty-six guinea pigs were randomly selected into four groups with 9 guinea pigs in each grup;group A(normal saline) ,group B(gentamicin) ,group C( gentamicin +.hyperbaric oxygen) and group D( gentamicin + hyperbaric air). Animals in group A were injected peritoneally with normal saline,and those in group B,C and D injected peritoneally with gentamicin of 150 mg/( kg · d). Animals in C were exposed to hyperbaric oxygen,and those in group D to hyperbaric air,both at 0.2 MPa once a day before the injection of gentamicin, consecutively for five days. The day before and five day after exposure, we assessed cochlear function of hearing in the animals was assessed by auditory brainstem response (ABR) test. Structural changes in cochlear hair cells were also examined by scanning electronic microscopy after the animals were dissected. Results The ABR threshold shifts were not significantly different between group C and group A (P 〉 0.05 ). In contrast,the ABR threshold shifts in group C were significantly different from those in groups B and D, respectively(P 〈0.01 ). The structural changes in the cochlear hair cells were consistent with the results from ABR test. Conclusions Pre-exposure to hyperbaric oxygen may have a short-term effect on the prevention of gentamicin-induced cochlear damage in guinea pigs.
