摘  要:目的探讨小檗碱预防颈动脉粥样硬化形成的作用机制。方法将32只雄性新西兰大白兔随机分成4组:正常组、颈动脉粥样硬化组(模型组)、生理盐水组(对照组)、小檗碱预防组(小檗碱组);除了正常组给予正常饮食外,其余各组行高脂饲料喂养加颈动脉内膜空气干燥术建立颈动脉粥样硬化模型,在此基础上对照组和小檗碱组分别肌注生理盐水和小檗碱,术后4周麻醉处死;取动脉血及颈动脉组织分别测定循环型和组织型血管紧张素Ⅰ转换酶(ACE)活性和血管紧张素Ⅱ(AngⅡ)含量,挑选部分颈动脉行HE染色作为模型阳性筛选。结果模型组血管病变以泡沫细胞为主,形成动脉粥样斑块;对照组血管病变是以泡沫细胞为主的动脉粥样硬化灶;小檗碱组血管主要显示内膜明显增厚;模型组组织型ACE活性和AngⅡ含量明显高于小檗碱组和正常组(P<0.05)。结论小檗碱可抑制ACE活性,特别是颈动脉ACE活性,降低组织中AngⅡ含量,预防颈动脉粥样硬化形成。Objective To explore the mechanism of the prevention of Berberine on the carotid artery atherosclerosis. Methods Thirty-two New Zealand white rabbits were divided into four groups: normal group, carotid artery atherosclerosis (model group), physiological saline group (control group), Berberine prevention group (Berberine group). Except normal group with normal diet, other group were performed with high cholesterol-diet and air-dry on the common carotid, on the basis of that, physiological saline group and Berberine group were treated with intramuscular physiological saline injection and Berberine respectively. After air-dry for four weeks, all rabbits were executed with anaesthesia. Arterial blood and air-dried carotid artery were collected for measuring angiotensin-converting enzyme (ACE) and angiotensin Ⅱ (Ang Ⅱ) of plasma and tissue. Parts of carotid artery were stained with HE method for choosing perfect sample. Results Model group was characterized with foam cells which formed atheromatous plapue, control group atherosclerosis lesion of foam cells chiefly, and Berberine group thickening intima. Compared with berberine group, model group was increased in tissue ACE, Ang Ⅱ and plasmic ACE obviously (P〈0.05). Compared with normal group, tissue ACE and Ang Ⅱ was increased in model group markedly (P〈0.05). Conclusions Berberine can deplete activity of ACE, especially carotid artery ACE, and decreased level of tissue Ang Ⅱ, so as to prevent development of carotid artery atherosclerosis.
