氟哌啶醇血浆浓度与治疗精神分裂症疗效的相关分析

摘　　要：为了进一步探讨氟哌啶醇（ＨＬ）血浆浓度与治疗精神分裂症疗效的关系，对病程小于５年的３０例精神分裂症患者，以ＨＬ０．２０ｍｇ·ｋｇ－１·ｄ固定剂量治疗６周，以放射免疫法测定ＨＬ、游离氟哌啶醇（ＦＨＬ）的血浆浓度。结果：血药浓度与临床疗效呈显著负相关，ＨＬ血浆浓度在２．７～２２．７μｇ／Ｌ，ＦＨＬ血浆浓度在０．２５～２．１１μｇ／Ｌ范围的患者临床疗效较好。提示ＨＬ治疗精神分裂症存在合宜血药浓度范围，同时提示在浓效关系的研究中。Theaimofthisstudywastoseethecorelationoftheclinicalresponseandtheplasmaconcentrationofhaloperidolinthetreatmentofschizophrenia.30schizophrenicpatientsweretreatedwithhaloperidolinthedosageof0.20mg·kg-1·dayfor6weeks.Haloperidol(HL)andfreehaloperidol(FHL)plasmalevelsweremeasuredwithradioimmunoassay.TheresultsshowedthattheclinicalresponsewerenegativelycorrelatedtotheplasmaconcentrationwhilethesideeffectsofHLtherapywerepositivelycorrelatedtoitsplasmalevel.Thepatientswhoseplasmalevelsrangedfrom2.7to22.7μg/LforHLor0.25to2.11μg/LforFHLyieldedbetteroutcomeattheendofthetreatment.Thestudysuggestedthattheoptimalplasmaconcentrationofhaloperidalachievedadesirableresultinthetreatmentofschizophrenia.ItalsodemonstratedthattheFHLplasmaconcentrationwouldbeamoresensitiveindicatorintheresearchofthecorrelationbetweenthedrugresponseandtheplasmaconcentrationofthedrug.
