摘 要:目的:观察大鼠实验性舌癌癌变过程中胎盘型谷胱甘肽S-转移酶(GST-P)的表达特点。方法:6周龄雄性SD大鼠42只,随机分为2组。实验组动物(n=30)的饮水中加入体积分数为0.001%的4-硝基喹啉-1-氧化物(4NQO)水溶液7mg/(kg·d)。对照组(n=12)则仅给予饮用水。实验第2周、4周、8周、12周、16周和26周末,处死大鼠,其中实验组各5只,对照组各2只。处死动物后,从会厌部至口底前缘整体分离舌体,观察其大体形态,常规组织处理,免疫组化法检测舌黏膜组织内GST-P。结果:对照组大鼠未出现任何舌黏膜病变,舌体标本均呈GST-P阴性。实验第8周末,实验组大鼠舌黏膜开始出现上皮异常增殖,实验第16周末即有1只大鼠发生舌癌,26周实验组大鼠均出现舌癌。2周时实验组大鼠舌黏膜内开始出现GST-P阳性细胞,随着时间的推移,GST-P阳性灶数目和大小逐渐增加。结论:GST-P可能与舌癌的发生有关。GST-P阳性反应早于大鼠舌黏膜的形态学改变,可作为反映癌前细胞代谢特点的良好标记。Aim : To evaluate the expression of placental glutathione S-transferase (GST-P) in lingual carcinoma tissue of rat induced by 4-nitroquinoline 1-oxide (4NQO). Methods:A total of 42 male SD rats ( six weeks) were allocated into 2 groups. The experimental group (n = 30) was administrated with 4NQO in drinking water. The control group (n = 12) was given drinking water. At the end of 2 weeks, 4 weeks, 8 weeks, 12 weeks, 16 weeks and 26 weeks, rats were executed. The expression of GST-P in the tongue epithelium was studied using immunohistochemistry. ReSults: Histologically, lin- gual epithelial dysplasia was detectable after 8 weeks of 4NQO treatment and the number of dysplastic lesions increased with time. Development of lingual carcinoma begun at the end of the 16th week of the experiment. All animals developed lingual carcinoma at the end of the 26th week. GST-P^+ loci occurred at the 2nd week of the experiment, earlier than the occurrence of morphologic changes of epithelial dysplasia. Both the number and size of GST-P^+ loci in the tongue mucosa increased with time. Conclusion: These results indicate that GST-P is an useful marker in rat lingual carcinogenesis.
