摘  要:目的:探讨雌激素对甲状腺癌细胞生长的影响及其作用机制。方法:采用原代细胞培养获得甲状腺乳头状癌细胞,用不同浓度的β-雌二醇与甲状腺乳头状癌细胞共培养,4-甲基偶氮四唑蓝(MTT)法观察甲状腺癌细胞增殖情况,流式细胞仪DNA定量法分析雌二醇对甲状腺癌细胞细胞周期的影响,半定量RT-PCR法测定雌激素对甲状腺癌细胞CyclinD1mRNA表达的影响。结果:β-雌二醇刺激甲状腺乳头状癌细胞的增殖,并具有时间和浓度依赖性(P<0.05);β-雌二醇刺激甲状腺癌细胞后G0-G1细胞的比例减少,S期细胞增加(P<0.05);CyclinD1mRNA的表达升高(P<0.05)。结论:雌二醇促进甲状腺癌细胞从G1期向S期的转化而促进甲状腺癌细胞生长。Objective : To investigate the effects of β-estrodiol on proliferation of thyroid papillary carcinoma cells, and to investigate to investigate the mechanisms possibly involved. Methods: The primary cultured papillary thyroid carcinoma cells were incubated with different concentrations of β-estrodiol and harvested at different times. The cell viabilities were measured through dimethyhhiazol-diphenyhetrazolium bromide (MTY) assay, and the cell cycles of the stimulated cells were analyzed by flow cytometer. The expression of cyclinD1 mRNA was observed by semi-quantitative RT-PCR. Results: β-estrodiol obviously promoted the proliferation of thyroid carcinoma cells in a dose-dependent and time-dependent pattern (P 〈 0.05), which was inhibited by estrogen antagonist tamoxifen. After the treatment of β-estrodiol,the proportion of cells in G0-G1 phase decreased and cells in S phase increased significantly (P 〈 0.05). The cyclinDl mRNA expression increased under 10-s mol/L β-estrodiol stimulation. Conclusion: β-estrodiol can promote the growth of papillary thyroid carcinoma cells by facilitating the G1/S phase transition.
