摘　　要：目的探讨上皮性卵巢癌组织中二肽基肽酶(DPP)mRNA的表达及临床意义。方法设计5′端含有T7RNA聚合酶启动子序列的PCR引物,以质粒pcD26His的cDNA为模板扩增得到DNA片段,并在T7RNA聚合酶作用下,采用体外转录方法合成FITC标记的DPP RNA探针,对86例卵巢上皮性癌石蜡包埋组织进行原位杂交,检测其DPP mRNA的表达,12例正常卵巢组织作对照。结果86例卵巢癌组织中的DPP mRNA0、1、2、3级表达者分别为2、18、40和26例,阳性表达率为97.67%(84/86),显著高于对照组的16.66%(2/12),P<0.01;其表达与卵巢癌组织学分级和临床分期有关。结论DPP mRNA在卵巢癌组织中的表达增高,在卵巢癌的发生发展中起一定作用。[Objective] To explore the expression and significance of dipeptidyl peptidase （DPP） IV mRNA in patients with epithelial ovarian cancer. [Methods] PCR was performed with gene-specific primer pairs, one containing T7 promoter sequence at the 5＇end to prepare template DNA for RNA transcription. Sense and antisense FITC-labeled RNA probes were generated by in vitro transcription. 86 formalin-fixed paraffin-embedded tissues were investigated for mRNA expression by in situ hybridisation with above probes. [Results] Of 86 patients with epithelial ovarian cancer, 97.67% （84/86） were positive stained for DPP ⅣmRNA, 16.66% （2/ 12） positive stainings were observed in control group, P d0.01. DPP Ⅳ mRNA expression was associated with histological grade and FIGO stage of ovarian cancer. [Conclusions] DPP Ⅳ is high and involved in the development of ovarian cancer.
