胡麻油烟凝聚物对人胚肺二倍体细胞增殖和DNA含量的影响

摘　　要：目的观察胡麻油烟凝聚物(SOFA)对人胚肺二倍体细胞增殖及DNA含量的影响,为研究胡麻油烟潜在致癌作用提供实验依据。方法用不同浓度(终浓度为25、501、00、200、400μg/mL)的SOFA染毒人胚肺二倍体细胞后,采用四甲基偶氮唑蓝(MTT)比色法观察SOFA对人胚肺二倍体细胞的毒性作用,流式细胞术DNA含量分析法检测细胞周期及DNA倍体指数(DI值)的变化,并计算细胞增殖指数PI值。结果在受试剂量范围内,SOFA对人胚肺二倍体细胞生长有明显的抑制作用,且呈时间-剂量依赖关系(r24h=-0.685,r48h=-0.734,r72h=-0.722,P<0.01;r=-0.430,P<0.01);流式细胞仪检测结果显示,SOFA对人胚肺二倍体细胞S期有促进作用,反映细胞增殖活力的细胞增殖指数增高,并表现出一定的剂量依赖性(r=0.914,P<0.01),DI值呈现异倍化趋势。结论SOFA能够抑制人胚肺二倍体细胞生长,对人胚肺二倍体细胞具有毒性作用;SOFA可诱发人胚肺二倍体细胞细胞周期紊乱,细胞异常增生,DNA含量异倍体化,具有潜在的致癌作用。Objective To study the influence of siritch oil fume aggregates（SOFA） on the proliferation and cell cycle of human embryo lung diploid cell. Methods The human embryo lung diploid cell were exposed to SOFA at varing concentrations of 25,50,100,200,40μg/mL. MTT was used to detect the toxicity of SOFA to the human embryo lung diploid cell. Analysis of cell DNA content was used to analyze cell cycle and DNA index by flow cytometry, and calculate the proliferous index. Results The SOFA within the dose range of the experiment （25 ～400μg/mL） could restrain the growth of human embryo lung diploid cell, and with an obvious doseresponse relationship（ r24h = -0.685, r48h = -0.734, r72h = -0.722, P 〈 0.01 ; r = 0.430, P 〈 0.01） ; The result of analysis cell DNA content indicate that SOFA could result in the promotion of S phase, the increase of prolifera- tion index（PI）, correlate with the exposure levels（ r = 0. 914, P 〈 0.01 ）, and DNA index is with the trend of aneuploid. Conclusion SOFA could inhibit the growth of human embryo lung diploid cell and have the toxicity to human embryo lung diploid cell. SOFA could induce cell pretematural hyperplasia, the cell cycle foul-up, DNA content aneuploid. SOFA has the function of potential carcinogenicity to human.
