摘  要:目的:探讨低氧条件下HIF-1α及P53在大鼠视网膜的表达及相关性。方法:将大鼠56只随机分为2组,每组各28只,分别饲养于常氧和50mL/L低氧仓。于饲养后0,2,6,8,12,24,48h各处死4只,分别进行光镜,电镜,免疫组化(SABC)测定HIF-1α及P53表达,并进行相关性分析。结果:HIF-1α在缺氧后2h开始表达,8h达到高峰,12~24h持续强表达,48h表达明显下降。P53在缺氧后2h开始表达,以后表达逐渐升高,24h后表达达到高峰,48h表达明显下降。HIF-1α及P53在大鼠视网膜表达呈明显正相关(R=0.902495)。电镜结果显示在视网膜缺氧6h视网膜超微结构开始变化,24h凋亡达到高峰。结论:低氧条件下HIF-1α及P53在大鼠视网膜均有表达,随着HIF-1α的表达达到高峰后P53的表达达到高峰,两者呈明显正相关。P53表达达到高峰时电镜结果显示凋亡达到高峰。AIM: To investigate the expression of HIF-1α and P53 after hypoxia and to find the correlation between them. METHODS: Animals were divided into two groups with 20 rats in each group, and then bred in normoxia and hypoxia room. Two animals were executed after breeding for 2, 6, 8, 12, 24 and 48hours. HE staining was done to observe the common change of the rat retinal slices and immunohistrochemical staining to detect the expression of HIF-1α and P53 protein in those slices. Through electron microscope we observed the ultrastructure of the retinal. RESULTS: HIF-1α started to express after 2-hour hypoxia and express step up gradually achieve to the peak after 8hours. The expression keeps on strong from 12 hours to 24hours, the expression decrease obviously after 48 hours. p53 started to express after 2-hour hypoxia and express step up gradually achieve to the peak after24 hours, the expression decrease obviously after 48hours. We can see the ultrastructure started to change after 6 hours, and apoptosis achieved to the peak after 24 hours. CONCLUSION: HIF-1α and P53 are expressed in rats' retinal under hypoxia condition. The expression of p53 arrives to the peak after HIF-1α . There is obviously positive correlation between HIF-1α and p53. The retinal apoptosis under hypoxia condition may be mediated by p53.
