摘  要:目的:研究siRNA对胃癌细胞BGC-823生长抑素(SOM)分泌的抑制效应。方法:设计4条针对SOM基因不同位点的寡核苷酸序列,应用RiboMAXT7体外转录合成siRNA并转染胃癌细胞系BGC-823,经RT- PCR、免疫细胞化学法检测SOM mRNA和蛋白的表达水平,筛选抑制效果最佳的序列,MTT法检测BGC-823细胞的增殖变化。结果:转染后24、48及72h,SOM基因的表达被抑制,抑制效率有差异,并呈浓度及时间依赖性。结论:体外合成siRNA抑制胃癌细胞系SOM基因的表达,增强了细胞的增殖能力。Objective: To study the inhibitory effect of various siRNAs on expression of somatostatin (SOM) in gastric cancer cell line BGC- 823. Methods: Four nucleotide sequences of siRNAs corresponding to various sites of somatostatin gene were designed, siRNA was synthesized by in vitro transcription RihoMAXT7 and was transfected into gastric cancer cell line BGC 823. The inhibitory effects were detected by RT-PCR and immunohistochemical techniques to select the best sequence that inhibited SOM gene expression. MTT assay was used to determine the growth of BGC 823 cells. Results: The somatostatin gene expression was suppressed at 24 h, 48 h, and 72 h after transfection and the efficiency was different, displayed in both a time-and concentration-dependent manner. Conclusion: siRNAs synthesized in vitro inhibit SOM gene expression in gastric cancer cell line, resul ring in promotion of BGC-823 cells proliferation. s
