摘 要:目的寻找促神经干细胞分裂增殖物质,为神经系统发育研究和神经系统疾病的治疗研究提供新的依据。方法用硫酸铵盐析及超滤离心法分离骨骼肌蛋白质,体外培养新生小鼠神经干细胞,通过神经球生成的观察、特异性蛋白质免疫细胞化学染色和MTT比色检测法鉴定神经干细胞并判断其增殖能力。结果在培养物中有大量神经球生成,且与对照组差异有显著性(P<0.01),神经干细胞的特异性蛋白质(Nestin)免疫细胞化学染色呈阳性。结论骨骼肌提取液可促进体外培养神经干细胞增殖分裂,且呈一定量效关系,其有效成分为分子量≤100 ku的蛋白质分子。Aim To discover NSC proliferation-promoting factor, which will contribute to study on nervous system development and treatment of nervous system disease. Methods Separate the proteins from skeletal muscle extract by using ammonium sulfate precipitation and ultra filtration. The NSC of newborn mice were cultured in vitro, neurospheres were observed, immunocytoehemical staining of characteristic protein and MTr assay were performed to identify the NSC and their proliferative properties. Results A great deal of neurospheres were formed in cultured cells, and their characteristic protein (Nestin) of NSC labeling were positive showed by immunocytochemical staining. Conclusions The skeletal muscle extract can increase the total number of NSC in vitro in a dose-dependent manner, its effective components are the proteins whose molecular weight is less than 100 ku.
