摘 要:目的观察不同剂量^(125)I粒子对人胃癌裸鼠移植瘤的杀伤作用及组织损伤。方法建立人胃癌BGC-823细胞裸鼠皮下移植瘤模型,随机分为对照组和实验组,将不同剂量(100、120、140 Gy)^(125)I粒子植于实验组。30、60 d:比较移植瘤抑瘤率,病理组织学、局部皮肤反应、裸鼠体重及白细胞计数。结果30 d,100、120、140 Gy组抑瘤率分别为51.93%、79.18%、90.22%,病理组织学反应程度多为RCRG2(45.83%),各实验组组间除120 Gy与140 Gy(P>0.05)组外,及分别与对照组比较,差异有统计学意义(P<0.05);局部皮肤反应各实验组与对照组比较,差异有统计学意义(P<0.05),各实验组组间差异无统计学意义(P>0.05)。60 d,各组抑瘤率分别为97.97%、100%、96.69%,病理组织学反应程度以RCRG1居多(62.5%),与对照组比较,差异均有统计学意义(P<0.05),各实验组组间差异无统计学意义(P>0.05);局部皮肤反应随剂量增高而加重,各实验组组间及其分别与对照组比较,差异有统计学意义(P<0.05)。结论不同剂量的^(125)I粒子在不同时间对人胃癌裸鼠皮下移植瘤有显著杀伤作用,但120Gy和140Gy组随剂量累积,损伤明显增加。提示100 Gy可能是治疗人胃癌裸鼠皮下移植瘤的有效剂量。Objective To study the lethal effect of different absorbed doses and time of interstitial implantation with ^125I seeds for treatment of the human gastric tumor xenografts in nude mice. Methods Animal model simulating human gastric carcinoma was established by subcutaneous implantation of cultured BGC-823 cell lines into nude mice. Mice used in experiments were randomly divided into 4 groups and treated with different doses and different time. Tumor volume, inhibitory rate, tumor regression, regional skin reaction,animal weight and number of white blood cells were evaluated. Results After ^125I seeds implantation for 30 days,the growth inhibitory rate in 100,120 and 140 Gy groups was 51.93%, 79.18 % and 90.22 % respectively. Most cases were presented with RCRG2 (45.83 % ). Theie was significant difference between control and different dose groups ( P 〈 0.05). Between different dose groups there was significant difference ( P 〈 0.05 ). There was significant difference in regional skin reaction between control and different dose groups (P 〈 0.05 ). After 60 days, the growth inhibitory rate in 100,120 and 140 Gy groups was 97.97 %, 100 % and 96.69 % respectively. A pathological complete response RCRG1 was seen in 15 cases (62.5 % ). There was significant difference between control and different dose groups (P〈 0.01 ). Among different dose groups there was no difference (P 〉 0.05). There was significant difference in regional skin reaction between control and different dose groups (P 〈 0.05 ). There was no difference ( P 〉 0.05 ) in weight and number of white blood cells after treatment among the groups. Conclusion Different doses of ^125I seeds had significant lethal effect on the human gastric tumour xenografts in nude mice at different time points. 100 Gy might be the effective dose for treatment of the human gastric tumour xenografts in nude mice.
