扇贝多肽促进骨髓基质细胞体外增殖作用的研究

摘　　要：目的研究扇贝多肽(polypeptides form chlamys farreri,PCF)促进骨髓基质细胞(Bone marrow stem cells,BM-SCs)修复的机制和体外培养作用的最适浓度。方法将成年兔骨髓基质细胞分离纯化后培养24 h后分成4组,扇贝多肽组(按用药浓度分3组:0.01 mg/L组为B组,0.1 mg/L为C组,1 mg/L为D组)和正常组为A组,采用相差显微镜观察,每日计数测定BMSCs的增殖曲线,24 h、72 h后用流式细胞仪测定S期的比例。结果0.01 mg/L浓度对BMSCs的增殖作用不明显,C组和D组在24 h和72 h后处于S期的BMSCs明显增多。10 d后药物组细胞计数明显高于对照组。其中C组和D组和对照组的倍增时间分别为6.2、4.9、8.4 d。两者相比,差异有显著性(P<0.01)。结论扇贝多肽能显著地促进BMSCs的增殖从而促进神经损伤的修复。Objective To study the mechanism of Polypeptides from chlarnys farrer （PCF） facilitating the bone marrow stem cells （BMSCs） regeneration and search its optimal concentration in vitro. Methods The BMSCs from adult rabbit had been isolated, purified and cultured for 24 hours, then these cells were divided four groups, including control group （ group A） and PCF group, which were further divided into three subgroup B , C and D according to various concentration of PCF. To count numbers of the BMSCs every day under phase contrast microscope and draw growth curve. The proportion of BMSCs in S phase were determined by flow cytometry at 24th and 72nd hour. Results PCF at the concentration of 0.01 mg/L. did not promote the BMSCs proliferation. When PCF concentration increased up to 0.1 mg/L to 1 rag/L, the numbers of BMSCSs in S phase were more increase than that of control group at 24th and 72nd hour. The BMSCs counting in PCF group were higher than that in control group on tenth day. The doubling time of cells in the group of PCF concentration at 0.1 mg/L, lmg/L and 0 mg/ 1.（ control group） were 6.2,4.9 and 8.4 days respectively. There were statistically significant difference. Conclusions PCF could promote BMSCs proliferation, thereby facilitate renovation of nerve injury.
