摘  要:目的 通过建立缺血再灌注的大脑中动脉闭塞大鼠模型,观察大鼠认知功能改变及盐酸多奈哌齐、西酞普兰对其认知功能障碍的影响。方法 36只SD大鼠雌雄各半.随机分为假手术组(A组),缺血90min再灌注盐水治疗组(B组)、盐酸奈哌齐治疗组(C组)和西酞普兰治疗组(D组)。采用ZeaLonga's线栓法制成年SD大鼠大脑中动脉闭塞(MCAO)模型。观察术后B、C、D组大鼠神经行为学评分.A组、B组术后14d四氮唑红(TTC)染色.以及各组水迷宫及听觉事件相关电位(P300)测试。结果 术后大鼠神经行为学评分都在2分以上。B组大鼠术后14dTTC染色的结果显示了模型的可靠性。定位航行试验中:在实验的2~5d.B组大鼠逃避潜伏期显著长于A组(P〈0.01);C组、D组大鼠逃避潜伏期显著短于B组(P〈0.01);实验的3~5d,C组大鼠逃避潜伏期显著短于D组(P〈0.05)。空间探索试验中:C组、D组的平台象限游泳的时间和总游泳时间(tP/tT)、平台象限距离和总距离之比(dP/dT)显著大于B组(P〈0.05);B组的dP/dT、tP/tT显著小于A组(P〈0.01);C组dP/dT、tP/tT显著大于D组(P〈0.05)。听觉事件相关电位(P300)中4组之间均存在明显差异(P〈0.01)。结论 大鼠大脑中动脉闭塞90min后再灌注能导致认知功能的下降;盐酸多奈哌齐和西酞普均能改善大鼠认知障碍,但前者的效果可能更好。Objective To observe the variation of cognition ability of the middle cerebral artery occlusion (MCAO) rats following of cerebral ischemia-reperfusion and treated by Donepezil or Cipramil. Methods 36 SD rats were randomly divided into the sham group (group A),the ischemia 90 rain and reperfusion treated with noereal sodlium group(group B),Donepezil group(group C) and Cipramil group (group D) . The models of MCAO were made by occluding the MCA with thread according to Longa's method . Behavioral assessment were evaluat- ed by neurological test. Brain tissue damage was detected 14 days after the surgery by triphenyl tetrazolium chloride(TTC) staining. Cognition ability was determined by morris-maze test and P300 test. Results The scores of neurological test of ischemia groups were all above two points. The results of TTC staining of infarct areas of normal sodium group ascertained the reliability of Longa's method. After the first day in place navigation test ,escape latency(EL) of group B was significant longer than group A. EL of group C and D was significant shorter than group B . EL of group C was significant shorter than group D after the third day. The ratio of dP/dT and tP/tT of group C was significant higher than group B and D in spatial probe test. The ratio of dP/dT and tP/tT of group A was significant higher than group B. Significant differences were detected between each group in P300 test. Conclusion The ischemia of 90 min and reperfusion induced by MCAO can result in cognitive impairment. Donepezil and Cipramil are shown to improve significantly the cognitive function and the former may be more efficiently.
