摘  要:目的探讨外源性p16基因导入对人原发性肝癌细胞迁移能力和VEGF及nm23表达的影响。方法细胞划痕法检测外源性p16基因导入对人肝癌细胞SMMC-7721迁移能力的影响;免疫细胞化学法分析外源性p16基因导入对VEGF表达的影响;Western blotting法分析外源性p16基因导入对nm23表达的影响。结果外源性p16基因导入人肝癌细胞SMMC-7721后,转染组细胞迁移能力较空载体对照组和空白对照组SMMC-7721细胞明显下降。外源性p16基因导入人肝癌细胞SMMC-7721后,转染组VEGF表达较空载体对照组和空白对照组SMMC-7721细胞减弱。外源性p16基因导入人肝癌细胞SMMC-7721后,转染组nm23表达较空载体对照组和空白对照组SMMC-7721细胞增强。结论外源性p16基因导入能降低人肝癌细胞SMMC-7721的侵袭和迁移能力。Objective To investigate the effect of exogenous p16 expression on migration ability, VEGF and nm23 expression of human hepatocellular carcinoma . Methods The migration abilities of SMMC - 7721 cell were tested using a scratchwound model in vitro. Protein level of vascular endothelial growth factor (VEGF) was assessed bv inununocytochemistry. Protein level of nm23 was assessed by Western blot. Results The migration abilities of SMMC - 7721 - p16 cells was obviously decreased compared with that of SMMC - 7721 - VECTOR and SMMC - 7721 cells. Protein level of vascular endothelial growth factor (VEGF) of SMMC - 7721 - p16 cells was decreased compared with that of SMMC - 7721 - VECTOR and SMMC - 7721 cells. Protein level of nm23 of SMMC - 7721 - p16 cells was increased compared with that of SMMC - 7721 - VECTOR and SMMC - 7721 cells. Conclusions Exogenous p16 expression can decrease the invasion and migration abilities of SMMC- 7721 cells.
