糖尿病大鼠肾脏PPARγmRNA表达及解毒通络保肾胶囊干预研究

摘　　要：目的:研究糖尿病大鼠肾脏组织过氧化物酶体增殖物激活受体γ(PPARγ)mRNA表达及中药解毒通络保肾胶囊的调节作用。方法:42只W istar大鼠用链脲佐菌素诱发糖尿病后,随机分为DM组、罗格列酮治疗组和解毒通络保肾胶囊治疗组,以正常10只大鼠作为对照。连续处理12周后,应用RT-PCR检测肾皮质过氧化物酶体增殖物激活受体γ(PPARγ)mRNA的表达水平。结果:DM大鼠肾皮质PPARγmRNA表达降低,解毒通络保肾胶囊治疗组PPARγmRNA高于DM组(P<0.05)。结论:解毒通络保肾胶囊对DM肾脏病变有保护作用,其机制可能是通过激活PPARγ途径,调控相关基因表达,改善糖脂代谢,减少细胞外基质的积聚。Objective： Study the expression of kidney cortex PPARγmRNA in DM big mouse and the jiedutongluobaoshen capsule＇ sadjusting action. Methods： 42 big mice were brought out DM after being dealed with STZ,than were divided into DM group, rosiglitazonetreated group and capsule treated group at random,and 10 normal mice were regarded as control group. Being treated after 12 weeks insuccession, measuring the expression level of kidney cortex PPARγwith PT - PCR. Results ： The expression of the DM big mouse kidney cortex PPARγwas reduced,the PPARγmRNA of the capsuletreated group was higher than DM group（ P〈0.05）. Conclusion： The jie du tongluo capsule has protective action on the kidney pathologicalchange caused by diabetes, its mechanism may be that adjusting andcontroling the expression of relevant genes through activating PPARγ, impoving the supersession of the glucose and fat and reducing thegathering of ECM.
