摘  要:目的研究siRNA对胃癌细胞BGC-823生长抑素(SOM)分泌的抑制效应。方法设计2条针对SOM基因不同位点的寡核苷酸序列,应用RiboMAXT7体外转录合成siRNA并转染胃癌细胞系BGC-823,经RT-PCR,免疫细胞化学法检测SOM mRNA和蛋白的表达水平,筛选抑制效果最佳的序列。结果转染后24 h、48 h及72 h,SOM基因的表达被抑制,抑制效率有差异。结论体外合成siRNA可以抑制SOM基因的表达,抑制效率呈现浓度及时间依赖性。Objective To study the inhibition effects of various siRNA on expression of somatostatin in gastric cancer cell line BGC-823. Methods Two nucleotide sequences of siRNA corresponding to different sites of somatostatin gene were designed, siRNA was synthesized in vitro transcription RiboMAXT7 and transfected into gastric cancer cell line BGC-823. The inhibitory effects were detected by RT-PCR and immunohistochemical techniques to select the best sequences that inhibit SOM gene expression. MTT assay was used to determine the growth of BGC-823 cells, Results The somatostatin gene expression was suppressed for 24 h, 48 h, and 72 h after transfection, and the efficiency was different. Conclusions siRNA was synthesized in vitro and it inhibited SOM gene expression, The inhibitory effects displayed in both a time- and concentration-dependent manner.
