摘 要:目的:研究大肠癌组织p27和Skp2蛋白表达,以及p27和Skp2表达与临床病理关系,探讨两者之间相关性。方法:采用免疫组织化学SP法检测30例大肠癌标本组织和18例正常大肠粘膜组织p27和Skp2蛋白表达。结果:p27在正常大肠粘膜组织中平均阳性率为55.2%,显著高于大肠癌组织(27.5%,P<0.05)。p27表达与大肠癌分化程度,Dukes'分期及淋巴结转移呈显著性负相关(P<0.05)。Skp2在大肠癌组织中平均阳性率为9.5%,显著高于正常大肠粘膜组织(1.8%,P<0.05)。Skp2表达与大肠癌分化程度呈显著性负相关(P<0.05),与患者年龄,性别,Dukes'分期及淋巴结转移无明显相关(P>0.05)。大肠癌中p27与Skp2表达呈负相关(r=-0.806,P<0.01)。结论:大肠癌中Skp2与p27降解有关,Skp2是大肠癌致癌基因,参与了大肠癌发生,可能成为大肠癌治疗新靶点。Objective: To investigate the expression and clinicopathologic characteristics of p27 and Skp2 proteins in colorectal carcinomas and to explore the relationship between p27 and Skp2 in colorectal carcinomas. Methods: The expression of p27 and Skp2 proteins were determined using immunohistochemical S-P method in 30 cases with colorectal carcinomas and 18 with normal colorectal tissues. Results: The average positive rate of p27 protein in normal colorectal tissues was 55.2%, which was significantly higher than that in colorectal carcinomas (27.5%) (P〈0.05). The p27 expression in colorectal carcinomas was significantly associated with Dukes stage, regional lymph node status and histological grade (P〈0.05). The average positive rate of Skp2 protein in colorectal carcinomas was 9.5%, which was higher than that in normal colorectal tissues (1.8%) (P〈0.05). The Skp2 expression was associated with histology in colorectal carcinomas (P〈0.05). However, there was no correlation between the Skp2 expression and age, sex, nodal involvement and Dukes stage (P〉0.05). There was a reverse relationship between the p27 expression and Skp2 expression in colorectal carcinomas (r=-0.806, P〈0.01). Conclusion: The Skp2 protein was involved in the degradation of p27 protein. Skp2 was a potential oncogenic gene and associated with the development of colorectal carcinoma. Skp2 could be a promising target for future cancer therapy strategies.
