摘  要:目的探讨细胞骨架蛋白Ezrin在胃癌中的表达及其意义。方法采用免疫组织化学方法(免疫组化法)检测组织芯片上90例胃癌组织及12例正常胃组织标本中Ezrin的表达,结合胃癌的临床病理特点进行分析。同时应用Western印记检测胃癌细胞株的Ezrin的表达,体外侵袭试验验证其侵袭能力。结果90例胃癌组织中Ezrin的阳性表达率为93.3%,而12例正常组织Ezrin表达均阴性,两者比较P<0.05,差异有统计学意义。同时,Ezrin阳性率与胃癌患者有无区域淋巴结转移相关(P<0.05)。胃癌MKN-45细胞株中Ezrin的表达(171.72)明显高于其他4个细胞株(47.44~132.51);同时,MKN-45细胞的侵袭能力(220.21)也强于其他4株胃癌细胞(89.23~210.35)。结论胃癌组织中的Ezrin阳性表达显著高于正常胃组织,并与胃癌的转移相关。Objective To investigate the expression of Ezrin in gastric cancer, its role in tumor metastasis. Methods Ezrin expression in tumor tissues from 90 gastric cancer cases and in normal gastric mucosa from 12 cases with benign disease was examined by immunohistochemical staining. Ezrin expression in gastric cancer cell lines was also detected by Western blot, and in vitro invasion assay was used to examine the invasive ability of the cell lines. Results The expression rate of Ezrin was significantly higher in gastric cancer tissues than that in normal tissues( P 〈 0. 05), and significantly correlated with lymph node metastasis ( P 〈 0.05 ). Western blot showed that MKN-45 cell line had the highest expression of Ezrin among 5 gastric cancer ceils. MKN-45 possessed highest invasion ability. Conclusion Ezrin expression is up-regulated, and may be associated with lymph node metastasis in gastric cancer.
