摘　　要：目的观察肿瘤坏死因子α(TNFα)对KM小鼠骨骼肌和肝细胞膜胰岛素受体酪氨酸蛋白激酶(TPK)活力的影响。方法80只清洁级KM小鼠随机分为TNFα高剂量连续注射组(TH组,n=20)、TNFα低剂量连续注射组(TL组,n=20)、TNFα高剂量一次注射组(T1组,n=20)和正常对照组(C组,n=20)。密度梯度离心分离组织细胞膜,麦胚凝集素(WGA)亲和柱层析纯化膜胰岛素受体,外源聚谷-酪多肽底物同位素32P掺入法测定酪氨酸蛋白激酶(TPK)活力,观察各组胰岛素刺激前后的离体骨骼肌和肝细胞膜胰岛素受体TPK活力变化。结果1)TH和TL组基础(无胰岛素刺激)骨骼肌和肝细胞膜胰岛素受体TPK活力均明显低于C组,且TH组明显低于TL组(P均<0.01);2)TH组和TL组胰岛素刺激的骨骼肌和肝细胞膜胰岛素受体TPK活力变化值均明显低于C组,且TH组明显低于TL组,(P均<0.01);3)T1组无论基础还是胰岛素刺激的骨骼肌和肝细胞膜胰岛素受体TPK活力变化值均与C组无显著差别(P>0.05)。结论TNFα可抑制组织细胞膜胰岛素受体TPK活力,在组织细胞膜胰岛素受体信号转导障碍中发挥重要作用。TNFα抑制组织细胞膜胰岛素受体TPK活力,需连续刺激,一次刺激作用效果不明显。Objective To study the effects of TNFα on activity of TPK（tyrosine protein kinase） of the membrane insulin receptor of the muscle and liver in KM mouse. Methods Eighty KM mouse were divided randomly to the TH group（TNFα high-dose group）, TL group（TNFα low-dose group）, T1 group（TNFα once-dose group） and C group（control group）.The activity of TPK of the membrane insulin receptor in the muscle and liver without and with insulin-stimulated in all groups were observed. Results 1）The activity of the TPK of the membrane insulin receptor in the muscle and liver without insulin-stimulated of TH and TL group were both lower significantly than that of C group, and TH group were lower than that of TL group.2） The changes of activity of the TPK of the membrane insulin receptor in the muscle and liver caused by insulin-stimulated of TH and TL group were both lower significantly than that of C group,and TH group were lower than that of TL group. 3）There were no significant difference of TPK of the membrane insulin receptor in the muscle and liver of the T1 group and C group. Conclusion TNFα can suppress the activity of TPK of the membrane insulin receptor in muscle and liver, and the effects of TNFα depends on the dose and time of TNFα treatment.
