摘  要:目的:观察人胃癌细胞系MGC-803,BGC-823对重组基底膜的侵袭作用,分析养胃抗瘤冲剂对其侵袭作用的影响及可能作用途径。方法:实验于2001-05/2001-10在中国中医研究院广安门医院肿瘤细胞生物学实验室完成。选取日本健康雌性大耳白兔21只,按随机数字表法分为7组,即养胃抗瘤冲剂组及拆方(益气方、活血方、解毒方、益气活血方、益气解毒方)组、空白血清组,制备含药血清。采用Transwell细胞培养小室建立人肿瘤细胞系体外侵袭模型,动态观察人胃癌细胞系MGC-803,BGC-823对重组基底膜的侵袭作用以及中药复方养胃抗瘤冲剂(由生黄芪、人参、白花蛇舌草、草河车、三七、苏木等组成,每克含生药3.22g,由中国中医研究院广安门医院制剂室提供)及拆方的含药血清、化疗药顺氯氨铂对胃癌细胞系侵袭作用的影响,侵袭抑制率(%)=(对照组侵袭细胞数-给药组侵袭细胞数)/对照组侵袭细胞数×100%,凡抑制率达30%以上,认为有抗侵袭作用。同时采用流式细胞仪通过单克隆抗体标记的方法检测侵袭转移相关黏附蛋白CD9,CD31,CD36,CD42a,CD44,CD49d,CD62p,CD41、凝血酶敏感蛋白及CD63的表达。结果:①各组人胃癌细胞系MGC-803,BGC-823对重组基底膜侵袭作用的抑制率比较:养胃抗瘤冲剂组、益气组、解毒组对人胃癌细胞MGC-803的抑制率均显著高于空白血清组(55.14%,32.43%,36.59%,0,P<0.05)。益气解毒、益气活血组作用优于单纯解毒、益气、活血组,但两组比较差异无显著性意义(44.86%,41.23%,P>0.05)。人胃癌细胞MGC-803与BGC-823比较,侵袭作用MGC-803强于BGC-823(空白血清组侵袭细胞数分别为60.75±4.57,56.35±3.25),养胃抗瘤冲剂及拆方对MGC-803的抑制作用强于BGC-823,但比较差异无显著性意义(P>0.05)。②各组人胃癌细胞系MGC-803,BGC-823侵袭转移相关黏附蛋白的表达水平比较:养胃抗瘤冲剂组肿瘤转移黏附蛋白CD63、凝血酶敏感�AIM: To observe the invasion potency of gastric cancer cell lines MGC- 803/BGC-823 to reconstitute basement membrane, and explore the effects of yangwei kangliu (YWKL) granule and its detach parts on the invasion and the correlated mechanisms. METHODS: The experiment was conducted in the Cancer Cell Biological Laboratory of Guang'anmen Hospital, China Academy of Traditional Chinese Medicine from May to October 2001. Twenty-one healthy female Japanese rabbits were randomly divided into 7 groups: YWKL group, detach parts groups (yiqi formula, huoxue formula, jiedu formula, yiqihuoxue formula and yiqijiedu formula) and normal control group. The rabbits were fed with traditional Chinese drug to prepare serum drug. Transwell chamber was used to establish the in vitro invasion model of human cancer cell lines to observe the invasion potency of gastric cancer cell lines MGC-803/ BGC-823 to reconstitute basement membrane and the effects of YWKL granule (composed of fresh milkvetch root, ginseng, oldenlandia, rhizoma, notoginseng, sappan wood and so on with 3.22 g crude drug per gram, provided by the Pharmaceutical Department of Guang'anmen Hospital, China Academy of Traditional Chinese Medicine) and detach parts, cisplatin on invasion of gastric cancer cell lines. The inhibitory rate of invasion (%)= (number of invasion cells in the control group-number in the treatment group)/number of invasion cells in the control group×100%, those with inhibitory rate of 30% or above were regarded having anti-invasion effect. The expression of adhesion molecule CD9, CD31, CD36, CD42a, CD44, CD49d, CD62p, CD41, thrombin-sensitive protein (TSP) and CD63 on MGC-803/BGC-823 surface was assessed by monoclonal antibody labeling method with flow cytometry.
