MT-3和bcl-2在膀胱移行细胞癌中的表达及临床意义

摘　　要：目的:探讨膀胱移行细胞癌(BTCC)中金属硫蛋白亚型(MT-3)的表达及其与bcl-2的相互关系。方法:采用逆转录多聚酶链式反应和原位杂交技术,对11例正常膀胱黏膜和23例BTCC进行MT-3和bcl-2分析。结果:11例正常膀胱黏膜中MT-3和bcl-2mRNA表达阳性率分别为0%(0/11)和18.2%(2/11),23例膀胱癌中MT-3和bcl-2mRNA表达阳性率分别为78.3%(18/23)和39.1%(9/23);在MT-3和bcl-2组正常膀胱组织与BTCC组织表达比较差异有统计学意义(P<0.05)。MT-3的表达与bcl-2的表达密切相关(P<0.05)。结论:MT-3的表达与BTCC的早期诊断相关,可能成为一种新的肿瘤标志物;MT-3与bcl-2呈负相关,可能通过下调bcl-2的表达参与细胞调亡。Objective：To investigate the role of MT-3 in transitional cell carcinonmas of bladder and the possible relation with bcl-2. Methods：Using RT PCR technique, the expressions of MT-3 mRNA in 11 specimens from normal bladder mucosa and 23 specimens from bladder transitional cell carcinoma were measured. Bcl-2 mRNA expression was determined by in situ hybridization. Results： MT 3 and bcl-2 mRNA positive expression rates were found 0 and 18.2% in normal bladder mucosa respectively. But it was 78. 3% and 39. 1% in 23 BTCC specimens respectively. The positive rate was significantly higher in BTCC specimens than that in normal tissue （ P〈0.05）. The expressions of MT-3 and bcl-2 mRNA were not related with the grade, stage and recurrence after postoperative intravesical chemotherapy. The expression of MT-3 mRNA was related with the expression of bcl-2 mRNA （ P〈0.05）. Conclusions：MT-3 mRNA doesn＇t exist in normal bladder mucosa, but bladder neoplasm has a positive MT-3 mRNA expression, which doesn＇t correlate with the degree of malignance. The expression of MT-3 was related with the expression of bcl-2 （ P〈0.05）. In addition, MT-3 may be involved in apoptosis.
