摘 要:目的研究中国北方人对非小细胞肺癌(NSCLC)的易感性及放疗敏感性与p53基因Msp I单核苷酸多态性(SNP)的关系。方法采用病例对照方法,通过合成序列特异性引物(PCR-RFLP)方法检测50例NSCLC患者及56例健康对照者的p53基因Msp I单核苷酸多态性位点的基因型。同时,观察不同基因型患者的放射治疗疗效,检测与辐射敏感性相关的基因型。结果NSCLC组的A1/A2杂和基因型频率明显高于对照组(χ2=4.38,P<0.05)。A1/A2杂合子患非小细胞肺癌的风险相对风险度为2.51(1.06<OR<5.96)。未观察到p53基因Msp I单核苷酸多态性与非小细胞肺癌病人的辐射敏感性有密切的相关关系。结论p53基因Msp I单核苷酸多态性位点的A1/A2杂和基因型为非小细胞肺癌的易感基因,但尚未发现该基因型与NSCLC的辐射敏感性相关。Objective To study the association between the susceptibility and radiation sensitivity of non-small-cell-lung cancer (NSCLC) and single nucleotide polymorphism(SNP) in Northern Chinese Population. Methods Using RFLP-PCR assays, p53 Msp Ⅰ genotyping was performed by amplifying DNA fragments with sequence specific primers and be digested by Msp I enzyme in 50 patients with NSCLC as well as 56 healthy controls. Results The A1/A2 genotype frequency was significantly higher in NSCLC patients than that of the healthy controls ( X^2 = 4.38, P 〈 0.05 ). The odds ratio of risk of A1/A2 hemozygotes in NSCLC patients was 2.51 (1.06〈OR〈5.96).Condusion In the Northern Chinese population,Msp Ⅰ A1/A2 genotype in 1353 gene is closely associated with the susceptibility of NSCLC. There is no significant relationship between Msp I SNP in 1353 gene and radiation sensitivity in NSCLC.
