摘 要:目的:探讨益髓生血灵对不同发育阶段小鼠珠蛋白表达的影响。方法:分别提取正常成年小鼠骨髓、孕鼠(孕21d)和胎鼠(孕14d)胎肝胎脾有核细胞总RNA,采用real-time PCR方法检测中药益髓生血灵干预后对不同发育阶段小鼠的βminor基因(类似于人γ-珠蛋白基因)表达的影响。结果:中药干预后,胎鼠(孕14d)的βminor珠蛋白基因的表达没有明显改变;孕鼠(孕21d)和正常成年鼠造血组织βminor珠蛋白基因的表达量明显提高,与对照组相比存在显著性差异(P<0.05,P<0.01)。结论:益髓生血灵治疗β-地中海贫血的分子机制之一,是可能改变珠蛋白程序性表达,开启在出生前本应关闭的γ-珠蛋白基因,增加HbF的合成而达到治疗效果。Objective: This paper intended to make an approach to the effect on expression of mice hemoglobin in different developmental stages treated with YiSuiShengXxueLing(YSSXL). Methods The total RNA of the marrow of normal adult mice and the nucleated cell within fetus liver and spleen in fetus mice (to be preguanted 21 days) and preguanted mice(to be preguanted 14 days) were respectively real-time PCR method was applied to detect the effect on gene expression of mice β^minor hemoglobin in different developmental stages treated with YiSuiShengXxueLing Results YSSXL had seldom effect on β^minor gene expression in fetus mice; but it has obviously up-regulated β^minor gene expression in hematopoietic tissue of pregnanted mice and normal adult mice ( P 〈 0.05, P 〈 0.01 ). Conclusion: One of the possible molecular mechanism of β thalassanemia therapy with YSSXL may be altering the procedural expression of hemoglobin, unlocking the γ-globin gene which should be close up prior to birth, increasing the HbF synthesis .
