摘  要:目的 测定野葛根及藤茎中的大豆苷元含量,探讨野葛藤的药用资源价值。方法 采用高效液相法建立测定大豆苷元含量的方法学,色谱条件Diamonsil C18色谱柱(4.6mm×250mm,5μm),流动相:甲醇-水(50:50),流速:1.0m1.min^-1,柱温:35℃恒温,检测波长:250nm。结果 大豆苷元在0.05~0.5μg·ml^-1范围内线性关系良好,野葛根和藤茎中所含成分相近,大豆苷元根含量与产地密切相关。结论 本法简便、准确、重现性好,为研究野葛藤组分含量提供了方法依据,客观反映了野葛植物的药用质量情况。OBJECTIVE To establish an HPLC method for the determination of daidein and mensurate the daidein in radix and vine of Pueraria lobata(wild. )ohwi. of different producing area. METHODS Diamonsil C18 column(4.6mm×250mm, 5μm) was used. The mobile phase was methanol-water (50 : 50). The flow rate was 1 ml·min^-1 and column temperature was set at 35℃. The UV detection wavelength was set at 250nm. RESULTS Daidein showed a good linear relationship at a range of 0.05~0.5μg·ml^-1 , r =0. 9999. The result showed that the content of daidein in radix and vine have some different, and have relation with producing area. CONCLUSION The method was simple, accurate, highly sensitive and reproducible, and offered a gist for study the component content in the vine of Pueraria lobata(wild. )ohwi.
