摘  要:目的探讨二烯丙基三硫(dially trisulfide,DATS)对人胃癌MGC-803细胞凋亡和凋亡调控基因Bcl-2和Bax的影响。方法用光学显微镜,荧光显微镜,流式细胞术检测凋亡,免疫组化研究人胃癌MGC-803细胞Bcl-2,Bax表达的变化。结果在光学显微镜及荧光显微镜下,通过DATS作用后的胃癌细胞出现了凋亡的典型形态学改变;流式细胞术检测可见亚G1峰出现。免疫组化显示,经DATS处理后,Bax的表达增强(P<0.05),Bcl-2表达未见明显改变。结论Bcl-2基因家族可能参与了DATS诱导的MGC-803细胞凋亡。Objective To study the effect of dially trisulfide(DATS) on apoptosis of MGC - 803 cells and apoptosis - regulated gene Bcl - 2 and Bax. Methods The changes of apoptosis and Bcl - 2/Bax gene expression were detected by light microscope, fluorescence microscope, flow cytometric analysis and immunohistochemistry. Result After MGC803 cells were exposed to DATS, the typical apoptotic morphological changes were observed under the light microscope and the fluorescence microscope. Flow cytometry analysis showed that treating MGC803 cells with DATS significantly increased the percentage of apoptosis cell. The protein expression of Bax was increased after DATS treatment. Conclusion Bcl- 2 gene family may participate in the DATS induced apoptosis of MGC - 803 cells.
