Determination of Nitrofuran Metabolites in Chicken Muscle by High Performance Liquid Chromatography-Tandem Mass Spectrometry

摘  要:建立了一种专属、灵敏的液相色谱-串联质谱法,同时检测鸡肌肉组织中硝基呋喃类代谢物:呋喃唑酮代谢物AOZ、呋喃它酮代谢物AMOZ,呋喃妥因代谢物AHD和呋喃西林代谢物SEM.鸡肌肉组织中硝基呋喃类代谢物经衍生化、提取后,采用电喷雾离子源,正离子检测方式进行分析.在0.5~20 ng/mL浓度范围内,4种硝基呋喃类代谢物均呈线性,相关系数r>0.99.在0.5、1和2 μg/kg 3种添加浓度,AOZ、AMOZ、AHD和SEM相应的平均回收率在80%~120%,RSD<16%.4种硝基呋喃类代谢物的检测限均可达到0.25 μg/kg.该方法专属性强、灵敏度高,适用于鸡肌肉组织中硝基呋喃类代谢物的检测.Developing a method for simultaneously determination of nitrofuran metabolites:AOZ (furazolidone' s), AMOZ(furahadone' s), SEM (nitrofurazone' s) and AHD(nitrofurantoin' s) in chicken muscle. The nitrofuran metabolites were derived and extracted from the chicken muscle, and detected by electrospray ionization and in the positive ion mode. The calibration curves were good linear between the ratio of the peak areas and concentrations of nitrofuran metabolites from 0.5 to 20 ng/mL,the correlation coefficient r 〉0.99. The average recoveries from spiked chicken muscle at the three concentrations of 0.5,1 and 2 μg/kg were from 80% to 120%, and RSD 〈 16%. The detection limits of nitrofuran metabolites were 0.25 μg/kg. This method is specific ,sensitive ,and suitable for determination of nitrofuran metabolites in the chicken muscle.
