摘 要:目的研究Caspase-1和NF-κb蛋白在糖尿病大鼠视网膜组织中的表达及氨基胍对其表达的影响,探讨糖尿病视网膜病变的发病机制。方法60只Wistar大鼠,8只作为正常对照组,其它用链脲佐菌素诱导糖尿病。糖尿病模型建立成功后,随机分组为糖尿病未治疗组(DM)和氨基胍治疗组(AG),其下又各设1月、3月、6月亚组。应用免疫组织化学法检测大鼠视网膜组织中Caspase-1和NF-κb蛋白的表达,及氨基胍对两者表达的影响。结果正常大鼠和DM1组视网膜无Caspase-1蛋白的表达,DM3组主要见于神经节细胞层;DM6组表达扩展到内核层和外核层。正常大鼠和DM1月组视网膜内外核层弱表达NF-κb。DM3表达波及神经节细胞层,DM6中NF-κb蛋白表达几乎遍及整个视网膜层。氨基胍治疗组中,Caspase-1和NF-κb的表达均有下降,其中AG3组与DM3组,AG6组与DM6组两者表达差别均有统计学意义(P<0.05)。结论早期糖尿病视网膜上Caspase-1和NF-κb的表达上调了,两者可能均参与了早期糖尿病视网膜病变的发生,其机制可能是导致了神经细胞的凋亡。氨基胍可通过抑制Caspase-1和NF-κb蛋白在视网膜上的表达来治疗糖尿病视网膜病变。Objective To study the expression of caspase-1 and NF-κb in the retina of diabetic rats and to investigate the possible pathogenesis of diabetic retinopathy. Methods 60 wistar rats,8 were used as control. Diabetes were induced on the other rats by the intraperitoneal injection of streptozotocin (STZ) at a doze of 60 mg/kg. Diabetes were characterized by an increase in blood glucose ( 〉 16.65mmol/L) 24 -48 hours after injection. The successful diabetic rat models were randomly divided into diabetic mellitus (DM) group and amino guanidine (AG) group. DM and AG groups were subdivided into DMI, DM3, DM6 and AGI, AG3, AG6 groups according to the duration of diabetes. The expression of Caspase-1 and NF-κb were studied by streptavidin - biotin complex (SABC) immunohistochemistry. And the effects of amino guanidine on the expression of Caspase-1 and NF-κ b were also studied. Result: no Caspase-1 positive cells were observed in the normal retina. In DMI group, no obvious expression of Caspase-1 were found, while in DM3 group, positive expression of Caspase-1 were found in ganglion cell layer(GCL). And in DM6 group, positive expression extend to the inner nuclear layer(INL) and the outer nuclear layer(ONL). The positive expression of NF-κb is low and only in INL and ONL of normal retina. It had no obvious change in DMI ,and gradually increased in DM3 and DM6 . That is ,the expression mainly in INL, ONL and GCL in DM3, and extend to almost the whole retina in DM6 . Under the intervention of amino guanidine ,the expression of Caspese-1 and NF-Kb were inhibited in each AG group. The distinction of positive expression between DM3 and AG3, DM6 and AG6 have statistical significance( P 〈 0.05 ) Conclusion: Diabetes elevated the expres sion of Caspase-1 and NF-κb, which may take part in the early events of the onset of diabetic retinopathy, the pathogenesis may be that Caspase-1 and NF-κb cause the apoptosis of retinal nerve cells.
