Effect of penehyclidine hydrochloride on cerebral infarction volume and behavioral parameters in rats with cerebral ischemia-reperfusion injury

摘  要:目的探讨选择性M胆碱受体阻滞剂(盐酸戊乙奎醚)对脑缺血再灌注损伤大鼠脑梗死体积和行为学变化的影响。方法雄性SD大鼠随机分为假手术组、缺血再灌注组、东莨菪碱组和盐酸戊乙奎醚组。参照文献建立三动脉阻断法全脑缺血模型,缺血前40 min分别腹腔注射生理盐水(1 ml)、东莨菪碱(0.01 mg/kg)和盐酸戊乙奎醚(0.01 mg/kg)。分别于脑缺血再灌注后1天、3天和7天用开阔法、平衡木法、攀绳和肌力试验测定其行为学指标,并取脑测定梗死体积。结果大鼠缺血再灌注后出现行为学异常,前脑散在梗死灶,但早期3组之间无明显差别。再灌注3天和7天,与缺血再灌注组比较,盐酸戊乙奎醚组和东莨菪碱组脑梗死体积较小,行为学指标改善。盐酸戊乙奎醚组较东莨菪碱组改善更加明显。结论盐酸戊乙奎醚能减少脑缺血再灌注损伤大鼠脑梗死体积的进一步扩大并且改善其行为学指标,此作用优于同等剂量的东莨菪碱。Objective To investigate the effect of a selective muscarinic receptor antagonist (penehyclidine hydroehloride) on cerebral infarotion volume and behavioral parameters in rat three - vessel occlusion models of acute global brain isehemia - reperfusion. Methods Seventy - two male SD rats were randomly divided into four groups: sham operated group, vehicle treated group (saline 1 ml,i.p, ), scopolamine treated group (0.01 mg/kg,i.p. ) and penehyclidine hydroehloride treated group (0. 01 mg/kg, i. p. ) with drugs injected 40 min before isehemia, respectively. The isehemic duration was 10 min. The motor activity tests (open field activity test, beam - walking test and grip test) were carried out, and subsequently, the. animals were saeririced for assessment of infarotion volume separately 1, 3 and 7 days after reperfusion. Results There was no evident distinction among the three isehemia- reperfusion groups 1 day after reperfusion. But on days 3 and 7, both penehyclidine hydroehloride treatment and scopolamine treatment reduced the cerebral infarotion volume ( P 〈 0.05 or P 〈 0.01 ) and improved the motor activity impairment (P〈0.05 or P〈0.01), as compared with the vehicle treatment. The improvement was more marked in penehyclidine hydroehloride treated group than in scopolamine treated group ( P 〈 0.05). Conclusion Penehyclidine hydrochloride may decrease the extent of cerebral infarction and improve the behavioral parameters in the rats with acute global isehemia- reperfusion injury, and these effects are better than that of the same - dose scopolamine.
