摘  要:目的研究白血病患者骨髓血管新生与Bcl-2蛋白关系。方法采用免疫组织化学法(EnVison)检测了33例白血病患者骨髓活检组织中Bcl-2蛋白的表达,用第Ⅷ因子抗体标记内皮细胞,计算微血管密度(microvesseldensity,MVD),同时采用ELISA法检测白血病患者外周血的VEGF含量。结果Bcl-2蛋白阳性表达、MVD、VEGF含量均明显高于对照组(P<0.05)。通过对这三者进行两两相关性分析,结果MVD与VEGF存在相关性(P<0.01),Bcl-2蛋白阳性率与VEGF存在相关性(P<0.05),但Bcl-2蛋白阳性率与MVD不存在明显相关性(P>0.05)。结论白血病患者骨髓Bcl-2蛋白阳性和血管新生的增加,可能在白血病的发展、治疗中有重要作用,相互促进白血病的进程。Objective To demonstrate the relationship between Bcl-2 protein and angiogenesis of the bone marrow in leukemia patients. Methods The surgical specimens from 33 leukemia patients were stained by immunohistochemical technique (EnVison) for Bcl-2 expression,while microvessel density was calculated by means of marking Vascular endothelial cells with antibody of Ⅷ factor. ELISA was employed to measure the level of VEGF in the blood. Results Ratio of positive Bcl-2 protein,MVD, VEGF in leukemia patients were significantly higher than those in normal control (P 〈 0.05). In the same time, their correlations were analyzed with leukemia patients. We found a direct correlation between the levels of VEGF and MVD (P 〈 0.01), a significant correlation between ratio of positive Bcl-2 protein and the levels of VEGF in the marrow of leukemia patients(P 〈 0.05). But there were no association between the ratio of positive Bcl-2 protein and MVD (P 〉0.05). Conclusion Bcl-2 protein and angiogene.sis of the bone marrow in leukemia patients are play an important role in the pathogenesis of leukemia. Altogether they might promote the development of leukemia.
