摘 要:目的:探讨低剂量辐射对裸鼠移植人胶质瘤U251细胞凋亡的影响。方法:35只荷人胶质瘤U251裸鼠,随机分为7个实验组(每组5只),即假照组(0mGy),D1组(75mGy),D2组(4Gy),D1-12h+D2组(D1照射后12h给予D2),D1-24h+D2组(D1照射后24h给予D2),D1-48h+D2组(D1照射后48h给予D2)和D1-72h+D2组(D1照射后72h给予D2)。各实验组以不同方式照射后4d,荷瘤裸鼠全部处死,采用TUNEL法检测裸小鼠移植人胶质瘤组织的细胞凋亡。结果:D1(75mGy)照射后,细胞凋亡指数(AI)增至(0·19±0·07),但与假照组(0·17±0·07)比较差异无显著性(P>0·05)。D2(4Gy)组、D1+D2各组细胞凋亡指数均明显增加,分别为0·54±0·05、0·64±0·04、0·63±0·04、0·65±0·11及0·67±0·07,与假照组比较差异均具有显著性(P<0·01)。与D2组比较,D1+D2各组细胞凋亡指数增加更为明显,两者比较差异均具显著性(P<0·05)。结论:75mGy照射对胶质瘤细胞凋亡可能有一定程度的影响,低剂量辐射与大剂量辐射有协同促胶质瘤细胞凋亡的作用。Objective To study the effects of low dose radiation (LDR) on apoptosis of transplantation tumor cells of human glioma (U251) -bearing nude mice. Methods Thirty-five tumor-bearing nude mice were divided into seven groups (n=5): sham-irradiated group, 0 mGy: D1 group, 75 mGy: D2 group, 4 Gy: D1-12 h+D2 group, the time between D1 and D2 was 12 hi D1-24 h+D2 group, the time between D1 and D2 was 24 h: D1-48 h+D2 group, the time between D1 and D2 was 48 h: D1-72 h+D2 group, the time between D1 and D2 was 72 h. The apoptosis of transplantation tumor cells of tumor (U251) - bearing nude mice was detected by TUNEL. Results There were TUNEL positive cells in all groups. The apoptotic index (AI) in 75 mGy group (0. 19±0.07) was slightly increased, but the difference was not significant compared with sham-irradiated group (0.17 ± 0.07) (P〉0.05). While in D2 and D1+D2 groups, the AI (0.54±0.05, 0.64±0.04, 0.63±0.04, 0.65±0.11, 0.67±0. 07) were increased, and the latter was more obviously. There were significant differences compared with sham-irradiated group (P〈0.01). Conclusion LDR may promote apoptosis of transplantation tumor cells to some extent , and could enhance high dose radiation to induce apoptosis of transplantation tumor cells.
