反相HPLC法测定血清中氯化水杨醛呋喃硫羰基腙合铜

摘  要:应用Shim-packCLC-ODS柱设计了测定家兔血清中新型杀菌剂氯化水杨醛呋喃硫羰基腙合铜浓度的反相HPLC方法。活性化合物的平均绝对和相对回收率分别为84.36%和99.80%;日内(n=5)和日间(n=15)的平均变异系数分别为4.89%和5.70%;最低检出浓度为0.02mg/L,线性范围为0.05~20mg/L(n=9,r=0.9999)。To determine the level of copper salicylaldehyde 2furanthiccarboxyhydrazonate dichloride(CSFTCHD) in serum of rabbits, a reversed-phase HPLC method has been developed by using Shim-Pack CLC-ODS collum (150mm×6mm id) . The mobile phase was a mixture of methanol and distilled water(70:30 ,v/v)and the eluent was monitored at 420 nm. The sample was extracted from serum into ethyl acetate and then the organic layer was evaporated under nitrogen gas. The residue was reconstituted in methanol and injected into chromatographic system and CSFTCHD eluted at 6.2min. The mean absolute and relative recovery were 84.36% and 99. 20%. The measurable lowest level was 0.02 mg/L and the standard curve was liner over the concentration range of 0. 05~20 mg/L(n= 9 , r=0.9999).
