摘 要:目的建立角膜血染动物模型并进行病理学观察。方法12只兔双眼角膜根据其血染达角膜后基质的1/3,2/3及全部基质分为角膜血染Ⅰ、Ⅱ、Ⅲ期组。损伤角膜内皮细胞及反复前房注血制作角膜血染模型。观察角膜血染过程,适时取角膜行苏木精-伊红(HE)染、普鲁士蓝染及透射电镜(TEM)检查。结果模型制作后平均2.2天3、.5天和7.4天分别形成角膜血染Ⅰ、Ⅱ、Ⅲ期,其HE片及普鲁士蓝染片中Ⅰ-Ⅲ期颗粒由少增多;普鲁士蓝染片中的蓝色颗粒出现在Ⅱ期的细胞内及Ⅲ期细胞内外;TEM观察到Ⅱ期即有细胞坏死,Ⅲ期有胶原溶解及成纤维细胞生成。结论本研究的角膜血染模型操作性强、重复性高、并发症少,与临床角膜血染的发生接近,可作为眼挫伤前房积血致角膜血染发病机制及药物干预研究的合适模型。Objective To establish an animal model of corneal blood staining in rabbit, and observe the pathologic features of it. Methods 24 eyes of 12 rabbits were designed for three groups correspending the 3 stages (Ⅰ、Ⅱ、Ⅲ) of development of cornea blood staining that the staining occupied 1/3, 2/3 and almost the whole stroma of cornea. Destructed the endothelial cells and injected of blood in the antefior chamber repeatedly. The corneas were observed daily and cut off when they had significant clinical appearances of each of the three stages for pathological examinations-HE, Perl' s Prussian blue staining and transmission electron microscopic (TEM) examination. Results In average 2.2,3.5 and 7.4 d formed Ⅰ、Ⅱ、Ⅲ stage of corneal blood staining respectively. Both HE staining and Perl's Prussian blue staining reflected that blue patti increased gradually among Ⅰ-Ⅲ stage. Perl' s Prussian blue staining disclosed intracellular hemoglobin particles in Ⅱ stage and extracellular as well as intracellular hemoglobin particles in Ⅲ stage in keratocyte. TEM examination showed the necorsis of keratocytes in hath Ⅱ and Ⅲ stages, and revealed the lysis of collagen fibers and the formation of fibroblast. Conclusions The rabbit model may be performed easily and repeatedly with less complications, and was similar to the clinical process of corneal blood staining. It can be useful for the study of pathogenesis and medical intervention of corneal blood staining.
