摘 要:目的:观察异丙酚预处理对全脑缺血再灌注大鼠脑组织S100B及神经元特异性烯醇化酶含量的影响,分析异丙酚的脑保护作用。方法:实验于2005-03/05在解放军广州军区武汉总医院动物实验中心完成,36只雄性SD大鼠,随机分为假手术组、脑缺血模型组和异丙酚预处理组,每组12只,异丙酚预处理组动物在缺血前腹腔注射异丙酚100mg/kg,采用线栓法制作全脑缺血模型,假手术组同样麻醉手术但不行颈总动脉夹闭术,脑缺血模型组麻醉后采用线栓法制作全脑缺血模型,3组动物于脑缺血再灌注后24h评估并记录动物的神经行为学评分,取大鼠脑组织行S100B和神经元特异性烯醇化酶测定。结果:36只大鼠顺利完成全部实验的有34只。脑缺血再灌注组和异丙酚预处理组各有1只动物在脑缺血再灌注后出现偏瘫无法进行余下实验,这可能与动物的个体差异不能耐受实验有关,随机取30例作为结果分析样本(每组10只)。①假手术组和异丙酚预处理组旷场评分、斜坡实验、悬挂时间评分均较脑缺血模型组高[(29.75±4.17),(24.25±7.51),(10.13±3.22)格;(8.13±2.36),(8.50±3.48),(16.25±3.79)s;(51.00±5.93),(32.75±6.13),(13.13±4.25)s;P<0.05~0.01];异丙酚预处理组悬挂时间评分较假手术组低(P<0.05),旷场评分和斜坡实验评分也低于假手术组,但差异无显著性意义(P>0.05)。②假手术组和异丙酚预处理组脑组织中S100B和神经元特异性烯醇化酶含量均明显低于脑缺血模型组[(0.35±0.07),(0.83±0.47),(1.47±0.88)μg/L;(4.35±1.24),(10.40±5.66),(19.68±9.73)μg/L;P<0.05~0.01]。假手术组和异丙酚预处理组比较差异有显著性意义(P<0.05)。结论:缺血前2h异丙酚预处理可以改善大鼠脑缺血再灌注损伤后的神经行为学评分,降低脑缺血损伤大鼠脑组织S100B及神经元特异性烯醇化酶的含量,减轻神经损害,有明显的脑保护作用。AIM: To study the effects of propofol pretreatment on SI00B and neurone specific enolase (NSE) of brain tissues in rats with cerebral isehemia-reperfusion, and analyze the protective effects of propofol on brain. METHODS: The experiment was conducted in the Experimental Animal Center of Wuhan General Hospital of Chinese PLA in Military Area from March to May 2005. Thirty-six SD male rats were randomly divided into 3 groups: sham-operation group, cerebral ischemia-reperfusion model group (model group) and propofol pretreatment group (pretreatment group) with 12 rats in each group. Rats in the pretreatment group were abdominally injected with propofol (100 mg/kg) before ischemia, and strand method was used to made models of cerebral ischemia. Rats in the sham-operation group were anesthetized without occlusion of carotid artery. Rats in the model group were made intocerebral ischemic models with strand method after anesthetization. Rats of three groups were evaluated at 24 hours after eerobral ischemic-reperfusion, and the neurological behavior of rats were recorded and scored. The S100B and NSE in brain tissues of rats were determined. RESULTS: Of 36 rats, 34 accomplished all experiments. In the model group and pretreatment group respectively, there was one rat suffered from semisideration after cerebral ischemic reperfusion, which might be related with individual endurance. Thirty rats were randomly selected to be the samples for analysis with 10 rats from each group. ①The scores of slope test, suspending time were higher in the sham-operation group and pretreatment group than those in the model group [(29.75±4.17), (24.25±7.51 ), (10.13±3.22) ; (8.13±2.36), (8.50±3.48), (16.25±3.79) s; (51.00±5.93), (32.75±6.13), (13.13±4.25) s;P 〈 0.05-0.01]. The score of suspending time in the pretreatment group was lower than that in the shamoperation group (P 〈 0.05), and the scores of open space and slope test were lower than those in the sham
