Hyperinsulinemic-Euglycemic Clamp in Conscious Rats Based on Tail Arterial and Vein Catheterization Technique

摘  要:目的利用尾动静脉插管技术实施清醒状态下大鼠高胰岛素-正糖钳夹实验并对该技术进行评价。方法正常喂养和高脂喂养W istar大鼠各5只,喂养4周后,清醒状态下鼠尾根局部麻醉,分别行尾动静脉插管,由尾静脉输入胰岛素(0.25 U.kg-1.h-1)和葡萄糖,同时尾动脉采血,进行高胰岛素-正糖钳夹实验。结果尾动静脉插管术后大鼠血糖在正常范围内,术后血糖无明显波动,无大鼠死亡;高脂喂养组大鼠的葡萄糖输注率显著低于对照组(P<0.01);血糖变异系数和葡萄糖输入速率变异系数分别为5.361%和14.273%。结论运用尾动静脉插管术进行清醒状态下大鼠的高胰岛素-正糖钳夹实验,安全可行且实验结果稳定、准确,具有操作简便、缩短实验周期等优点。Objective To utilize tail arterial and vein catheterization technique to perform hyperinsulinemic-euglycemic clamp in conscious rats and to evaluate this technique. Methods Wistar rats were fed with standard chow diet or high-fat diet (two groups, and five rats in each group). Four weeks later, conscious rats received local anesthesia on the tail root, then tail arterial and vein catheterization, respectively. Tail vein was used to inject insulin ( the infusion speed was 0.25 U· kg^- 1· h ^- 1 ) and tail arterial to obtain blood sample for performing hyperinsulinemic- euglycemic clamp. Results After tail arterial and vein catheterization, the blood glucose of the rats was in the normal range without significant fluctuation, and no death of rats was reported. The glucose infusion rate (GIR) of highfat diet group was significantly lower than that of control group (P 〈 0.01 ). The blood glucose coefficient of variation and GIR coefficient of variation were 5. 361% and 14. 273% , respectively. Conclusion Compared with routine jugular vein and carotid artery catheterization technique, hyperinsulinemic-euglycemic clamp in conscious rats based on tail arterial and vein catheterization technique is easy to perform, and has a shortening experimental period and low death rate of rats.
