摘 要:目的:比较五种甲硫氨酸(M et)相关培养液对胃癌细胞增殖活性的影响。方法:以左旋M et(L-M et)培养液组为对照组,用去除M et和同型半胱氨酸(Hcy)(M et-Hcy-)、去除M et并添加Hcy(M et-Hcy+)、以右旋M et(D-M et)替代L-M et、左旋和右旋M et(DL-M et)培养液培养胃癌SUN-16细胞,采用MTT法检测其24、48、72、96和120 h的吸光度(A490)值。结果:M et-Hcy-、M et-Hcy+、D-M et和DL-M et组中胃癌细胞的A490均显著低于对照组(P<0.01),且前三组间的A490接近,DL-M et组的A490界于其余三组和对照组之间;各实验组与对照组间的差异随时间延长而加大。结论:M et-Hcy-、M et-Hcy+和D-M et培养环境抑制胃癌细胞增殖的作用相似,DL-M et环境可造成M et半饥饿状态而抑制胃癌细胞,但效果较差;D-M et环境是目前造成肿瘤细胞M et饥饿的理想方法。Objective: To compare the effect of 5 methionine-related media on inhibition of gastric cancer cells' proliferation. Methods:Gastric cancer cell line SUN-16 was cultured with L-Met, Met^- Hey^-, Met^-Hey^+ , D-Met, and DL-Met media respectively. The group of L-Met medium was control. A490 was detected by MTT in 24, 48, 72, 96, 120 h. Results : Comparing with control group, A490 of SUN-16 cells significantly decreased in Met^- Hey ^- , Met^- Hey^+, D-Met, DL-Met media respectively( P 〈 0.01 ). It was similar to each other for Met^- Hey^-, Met^- Hey ^+ , D-Met medium to inhibit gastric cancer cells' proliferation, and they were superior to DL-Met medium. This effect was time dependency. Conclusion: Met^- Hey^-, Met^-Hey^+ or D-Met medium are similar in inhibiting gastric cancer cells. DL-Met medium can inhibit gastric cancer cells by methionine semi-starvation, but its effect is inferior to Met^- Hey^-, Met^-Hey^+ or D-Met medium. D-Met environment is the best method to make Met starvation.
