摘  要:目的:建立应用自动磁性细胞分选器(autoMACS)分选人CD4+CD2+5免疫调节T细胞(Tregs)的方法。方法:应用密度梯度离心法分离人外周血单个核细胞,应用autoMACS分选其中的CD4+CD2+5T细胞,流式细胞技术检测分选细胞的纯度。结果:分别从5.7×107及5.6×107外周血单个核细胞中分选出6.9×105及6.7×105个CD4+CD2+5T细胞,其纯度>98%。结论:应用autoMACS可高效分选出高纯度的CD4+CD25+T细胞,为深入研究其功能打下了基础。Objective : To establish a method of isolating human CD4^+ CD25^+ T cells with auto magnetic activated cell sorting (autoMACS) . Methods : The peripheral blood mononuclear cells (PBMC) of 2 health donors were isolated by density gradient centrifugation, then the CD4^+ CD25^+ T cells were selected by autoMACS and their purity were examined by flow cytometry. Results: 6.9×10^5 and 6.7 ×10^5 CD4^+ CD25^+ T cells which purity were higher than 98% were isolated from 5.7×10^7 and 5.6×10^7 PBMCs. Conclusion : High purity human CD4^+ CD25^+ T cells can be isolated effectively by autoMACS.
