大肠癌中血管内皮生长因子C及其受体3表达的研究

摘　　要：目的研究大肠癌中血管内皮生长因子C(VEGF-C)与其受体3(VEGFR-3)的表达,及其与大肠癌Duke’s分期之间的关系。方法随机选择临床确诊的47例大肠癌患者,对其进行Duke’s分期。并对经手术切除的大肠癌组织进行免疫组织化学染色,分别记录VEGF-C与VEGFR-3的标记分数。结果47例不同Duke’s分期的大肠癌组织标本中淋巴管生长因子VEGF-C的标记分数分别为：B期1．38±1．15,C期2．77±1．69,D期3．94±2．36；VEGFR-3的标记分数分别为：B期1．06±1．00,C期2．00±1．58,D期3．44±2．33；均随肿瘤分期的增加而升高,B、C、D期间具有显著性差异(P＜0．05)。VEGF-C的阳性表达率分别为：B期62．5%,C期84．6%,D期94．4%,随肿瘤分期的增加而升高,B期与D期之间具有显著性差异(P＜0．05)。不同Duke’s分期的大肠癌组织标本对VEGFR-3阳性表达率分别为：B期62．5%,C期84．6%,D期88．9%,随肿瘤分期的增加而升高,但无显著性差异。结论淋巴管生长因子VEGF-C及其受体VEGFR-3在大肠癌组织中高表达,其阳性表达率与大肠癌的Duke’s分期呈明显正相关,对肿瘤的浸润及淋巴结转移和疾病预后的评估有一定价值。并可能成为肿瘤治疗的新靶位点。Objective To determine the expression of vascular endothelial growth factor-C （VEGF-C） and its re ceptor-3 （VEGFR-3） in human colorectal carcinoma and to analysis their relationship with Duke＇s stage. Methods Forty-seven patients with colorectal carcinoma were studied, and were classified based on Duke＇s stage system. The expressions of VEGF-C and VEGFR-3 were examinated by immunohistochemical staining of colorectal cancer tissue. The expressions of VEGF-C and VEGFR-3 were recorded by the labeling score under microscope. Results The labeling score of VEGF-C at Duke＇s stage B, C, and D was 1.38±1.15, 2.77±1.69, and 3.94±2.36 respectively. The score of VEG- FR-3 at stage B, C, and D was 1.06±1.00, 2.00±1.58,and 3.44±2.33 respectively. The scores were increased with the advance of clinical stage. There was a statistical difference among stage B, C and D（P〈0.05）. The rate of VEGF-C expression at stage B was 62.5%, stage C 84.6%, and stage D 94.4% respectively. There was statistically significant difference between stage B and D（P〈0.05）. The rate of VEGFR-3 expression at stage B was 62.5%,stage C 84.6% ,stage D 88.9%, respectively. The rate of VEGFR-3 expression increased slightly with the advance of clinical stage. There was no statistical difference among them. Conclusions The high expression level of pro-lymphangiogenesis factor VEGF-C and its receptor VEGFR-3 has significant relationship with Duke＇s stage. VEGF-C and VEGFR-3 play important roles in colorectal cancer lymphatic metastasis. It may be a new target in the treatment of colorectal carcinoma.
