摘 要:目的探讨川芎嗪对哮喘大鼠白细胞介素-4(IL-4)、白细胞介素-13(IL-13)水平的影响及其防治哮喘的作用机制。方法60只SD大鼠随机分为正常对照组、哮喘模型组、地塞米松组、小剂量及大剂量川芎嗪组、联合用药组,以卵蛋白致敏和激发制备大鼠哮喘模型,用酶联免疫吸附试验(ELISA)检测支气管肺泡灌洗液(BALF)中IL-4、IL-13的水平。结果哮喘模型组BALF中IL-4、IL-13水平均高于正常对照组(P<0.001);地塞米松组和川芎嗪治疗各组均显著低于哮喘模型组(P<0.001);川芎嗪大剂量组和联合用药组降低IL-4和IL-13的作用与地塞米松组无显著性差异(P>0.05);川芎嗪大剂量组降低IL-4的作用与川芎嗪小剂量组无显著性差异(P>0.05),川芎嗪大剂量组降低IL-13的作用优于川芎嗪小剂量组(P<0.05);BALF中IL-4和IL-13水平呈显著正相关(r=0.714,P<0.01)。结论川芎嗪通过抑制IL-4和IL-13的水平而抑制哮喘气道炎症。Objective To explore the changes of the levels of interleukin-4 ( IL-4 ), interleukin-13 ( IL-13 ) in asthmatic rats and to evaluate the influence of ligustrazine on them. Methods Sixty male SD rats were randomly divided into 6 experimental groups ( 10 in each) : normal control group, asthmatic model group, desamethasone group, low-dose of ligustrazine group, high-dose of ligustrazine group, combinative therapy group. The asthmatic model was established by repeated inhalation of ovalbulium. The levels of IL-4 and IL-13 in bronchoalveolar fluid (BALF) were measured by ELISA. Results The IL-4 and IL-13 levels were much higher(P 〈 0. 001 ) in BALF in asthmatic model group than those in normal control group. The IL-4 and IL-13 levels in each treatment group were significantly lower than those in asthmatic model group (P 〈 0.05 ) , but were higher than those in normal control group ( P 〈 0.05 ) , but the levels in high-dose of ligustrazine group and combinative therapy group had no significant difference with those in desamethasone group(P 〉0.05). The IL-4 level in BALF in high-dose of ligustrazine group had no significant difference with that in low-dose of ligustrazine group ( P 〉 0. 05 ). The IL-13 level in BALF in high-dose of ligustrazine group was lower than that in low-dose of ligustrazine group(P 〈0. 05 ). A positive correlation between IL-4 and IL-13 levels in BALF was demonstrated(r = 0. 714, P 〈 0. 01 ). Conclusion ligustrazine can downregulate the levels of IL-4 and IL-13 in asthmatic rats and inhibit the airway inflammation.
