摘  要:目的观察黄芩素与辅酶Q10对实验性大鼠视网膜光损伤的防护作用。方法SD大鼠通过24h持续光照射,建立视网膜光损伤模型。于光照前24h及光照前30min两次尾静脉注射给药。观察视网膜组织病理学改变及流式细胞术检测视网膜细胞凋亡率。结果组织病理学结果显示,阳性对照组视网膜组织结构破坏严重,而黄芩素组、辅酶Q10组和联合用药组结构损伤明显减轻。流式细胞术检测结果,所有用药组视网膜细胞凋亡率明显低于阳性对照组(P<0·05),联合用药组对细胞凋亡率的降低作用明显优于两药单独用药组(P<0·05)。结论黄芩素与辅酶Q10对视网膜光损伤诱发的细胞凋亡具有保护作用和明显的协同抗凋亡作用。Objective Free radical scavengers has been demonstrated to ameliorate light-induced retinal degeneration. We introduced two free radical scavengers in photic injury model to explore the protective effect of baicalein and coenzyme Q10 on light-induced retinal degeneration in experimental rats. Methods Forty-five Sprague-Dawley (SD) rats were divided randomly into normal control group, positive control group, baicalein group, coenzyme Q10 group and baicalein combined coenzyme Q10 group. The photic damage model of SD rats was established by exposing to strong green fluorescent light with an illuminance level of (2 000± 120) Lux for 24 hours. The drugs were intravenouly injected 24 and 0. 5 hours before light exposure in different drug group ,separately. The rats in normal group did not received green fluorescent light, and the rats from positive control group only underwent green fluorescent light without the administration of drug. The changes of retinal histopathology were observed. Cell apoptosis of retina was detected by Annexin-V-fluorescein isothiocyanate/Propidium iodium labelling and Flow cytometry. Results In positive control group,the outer and inner segment of photoreceptor arranged disorderly and the outer nuclear layer was scarce and scattered, however, no obvious retinal alteration was seen in baicalein,coenzyme Q10 and combination group. The cell apoptosis ratios of retina were significantly decreased in baicalein group,coenzyme Q10 group and combination group (14.09 ±1.58%,12.43 ±2.25% and 8. 10±1.75%) compared with positive control group (25.83 ±2.92 %)(P〈0. 05),and the decrease of apoptosis ratio was more dominant in combination group ( P 〈0. 05 ). Conclusion Baicalein and coenzyme Q10 have a synergic protection effect on cell apoptosis of retina induced by photic injury.
