山萘黄素对人白血病细胞系HL-60体外增殖的抑制作用

摘　　要：目的观察山萘黄素对H L-60细胞体外增殖的抑制作用及其对细胞周期的影响。方法分别采用台盘蓝拒染法及流式细胞术,在不同条件下测定细胞增殖的抑制率及细胞周期的分布。结果应用台盘蓝拒染法,山萘黄素能够显著地抑制H L-60细胞的体外增殖并且呈现时效及量效关系。流式细胞术的结果表明:山萘黄素作用2h可使HL-60细胞的细胞周期分别阻滞于G2/M期和G0/G1期。结论山萘黄素能够明显抑制HL-60细胞体外增殖,影响其细胞周期分布。Objective To observed the effect of kaempferol on proliferation and cell cycle of human leukemic cell HL-60 in vitro. Methods The cell growth was tested by trypan blue stain assay and the cell cycle was analyzed by flow cytometry in the various conditions. Results Using the trypan blue stain assay we found that kaempferol could strongly inhibited the proliferation of HL-60 cell, the higher dose, the longer time and the more obvious effects. Flow cytometry analysis showed that kaempferol could arrest cell cycle progression in G2/M and G0/G1 phase in two hours, respectively, Conclusions Kaempferol is an effective flavonoid compounds which can inhibit the proliferation and can affect the cell cycle of HL-60 cell in vitro.
