摘  要:目的观察不同染毒剂量二甲基甲酰胺(DMF)对正常成人离体肝细胞的增殖抑制作用。方法用MTT比色法观察细胞生长变化,分别以1.56mmol/L、6.25mmol/L、25mmol/L、100mmol/L DMF为染毒剂量,以6h、12h、24h为染毒时间对人肝细胞进行培养。比较在不同时间下,不同染毒剂量组与阴性组的细胞生长率。结果同一染毒时间组内,不同剂量组的细胞生长变化率结果差异有显著意义(F=27.41、23.86、111.22,P<0.01),以100mmol/L DMF组细胞增殖率为最低,但不同染毒剂量组细胞生长变化率未见明显剂量-效应关系,染毒6h和12h 25mmol/L组的细胞生长变化率高于其他浓度组;同一染毒剂量组内,各不同染毒时间组的细胞生长变化率结果差异有显著意义(F=27.80、29.79、42.47、28.88,P<0.01),以24h组细胞增殖率为最低。结论与阴性组比较,DMF对细胞增殖有抑制作用;但不同染毒剂量组细胞生长变化率的剂量-效应关系不明显;随染毒时间延长,同一染毒剂量组内细胞增殖活性变化率有下降趋势,有一定染毒时间-效应关系。Objective To explore the inhibitory effect on cell proliferation of human hepatecyte by N, N-dimethylformamide (DMF) in vitro. Methods The human hepatecytes were exposed to DMF with dose of0mmol/L, 1.56mmol/L, 6.25mmol/L, 25mmol/L and 100mmol/L respectively and each dose group was exposed to DMF for 6h, 12h and 24h. Methyhhiazohetrazolium assay (MTr assay) was performed to compare cell proliferation rate among these groups. Results At same point of exposure time, cell proliferation rates among different dose groups had statistically significant difference ( P 〈 0.01 ), of which 100mmol/ L DMF group was lowest. But the dose-effect relationship could not be observed. The hepatecyte proliferation rates in 25mmol/ L DMF group with exposure time for 6h and 12h were higher than other groups. In same dose groups, cell proliferation rates among different exposure time groups had statistically significant difference ( P 〈 0.01 ), of which exposure for 24h was lowest. Conclusions Compared with control group, DMF has inhibitory effect on cell proliferation. However, a dose-dependent relationship between cell proliferation and DMF concentration is not obvious. Cell proliferation rate declines with the prolongation of exposure time to DMF, and time-effect relationship exists.
