摘  要:建立了青鱼分割物亚临界水解产物中氨基酸的HPLC测定方法,以DNFB为衍生试剂,在碱性范围内,60℃反应1h。:Hypeoil ODS-C18色谱柱(100mm×4.6mmi.d.,5μm),流动相为乙酸钠溶液及乙腈-水溶液,采用线性梯度洗脱程序。检测波长为360nm;柱温为45℃。回收率为90.40%~110.80%,精密度为0.91%~8.58%。17种常见氨基酸在55min内得到较好的分离,并测定了青鱼分割物水解液中的天冬氨酸、甘氨酸、丙氨酸、亮氨酸等14种氨基酸的含量。The HPLC analysis of amino acids in subcrtical hydrolysate of the fish solid waste was set up. In this method, DNFB was used as derivatizing agent. The derivatizing reaction conditions were: pH 〉 7, reaction temperature 60℃, and reaction time 1 h. The HPLC analysis conditions were:chromatographic column hypeoil ODS-C18( 100 mm×4. 6 mm i. d. , 5 μm) ; mobile phase: sodium acetate and acetonitrile-water ; detection wavelength 360 ran, column temperature 45℃ ; linear gradient elution program. The recovery rate was 90. 40% - 110. 80% and C, was 0. 91% - 8.58%. Under these conditions,17 kinds of amino acids were separated completely in 55 minutes and 14 kinds of amino acids such as Asp, Glu, Ala, Leu and so on in the hydrolysate from fish protein hydrolysis in subcritical water were determined.
