摘  要:目的观察戊四氮(PTZ)对神经元样PC12细胞内钙离子(Ca2+)浓度和核因子κB(NF-κB)活化的影响,进一步探讨癫的发病机制。方法将传代培养的神经元样PC12细胞随机分为PTZ干预的PTZ组和不经PTZ干预的对照组,运用免疫荧光法检测神经元样PC12细胞内的Ca2+浓度和NF-κB核区与胞浆区的比值(FN/FC)。结果PTZ组PC12细胞NF-κBFN/FC为0.8821±0.0077,明显高于对照组的0.2161±0.0267(P<0.05),提示NF-κB活化;PTZ组PC12细胞内Ca2+浓度的荧光值(213.45±6.44)明显高于对照组的(80.73±1.01)(P<0.05)。结论PTZ能够使神经元样PC12细胞内Ca2+浓度升高和NF-κB激活。NF-κB激活可能是癫疒间形成的机制之一。Objective To investigate the epileptogenesis by observing effects of pentylenetetrazol (PTZ) on intracellular Ca2^+ concentration and the activation of nuclear factor κappa B (NF-κB) in neuron-like PC12 cells. Methods The subcuhured neuron-like PC12 cells were divided randomly into PTZ group treated with PTZ and control group treated without PTZ. The changes of the concentration of intracellular Ca2^+ and the expression ratio of NF-κB in nucleus and in cytoplasm (FN/FC) were measured by immunofluorescence assay. Results The relative value of FN/FC of NF-κB in PTZ group (0.8821 ±0. 0077 ) was higher than that in control group (0.2161 ±0. 0267) in neuron-like PC12 cells ( P 〈 0. 05 ). The concentration of intracellular Ca2^+ in PTZ group ( 213.45 ±6. 44) was higher than that in control group ( 80.73 ± 1.01, P 〈 0. 05 ). Conclusions PTZ can increases the concentration of intracellular Ca2^+ and activate NF-κB in neuron-like PC12 cells. The activation of NF-κB may play an important role in epileptogeneses.
