针灸大鼠血清诱导人卵巢癌细胞株SKOV3细胞凋亡及相关因子的研究

摘　　要：目的:观察针灸大鼠血清对人卵巢癌细胞株SKOV3增殖和凋亡及相关因子Fas的影响。方法:针灸剌激Wister大鼠双侧足三里、三阴交穴,每日2次,连续6天,取大鼠"针灸血清"分别作用于SKOV3细胞,MTT法检测细胞的生长,48h后流式细胞术检测细胞凋亡率及Fas表达率。结果:Fas表达率明显增高,SKOV3细胞生长受到明显抑制,作用48h后的对照组、艾灸组和电针组细胞凋亡率分别为1.17%、9.36%和5.12%。结论:针灸血清可能通过增加Fas的表达诱导SKOV3细胞凋亡。Objective ：To observe the effect on the proliferation and apoptosis and apoptosis related factor Fas of human ovary cancer cell line SKOV3 treated with acupunctural and moxibustional serum. Methods-Zusanli and Sanyinjiao in both sides of Wister mice were acupunctured and moxibusted for 6 days and serum was collected. SKOV3 cell was treated with the serum. The cytoxicity was detected with MTT, and apoptosis and Fas expression of SKOV3 cell was detected with cytometry 48 hours after treated with mice serum. Results： The proliferation of SKOV3 cell was inhibited remarkably and the expression of Fas was increased significantly. The apoptosic rate were 1. 17%, 5. 12% and 9.36% in control, moxibustional and acupuncture group respectively. Conclusion：Acupunctural and moxibustional serum could inhibit proliferation of SKOV3 cell and induce its apoptosis by increasing the expression rate of Fas.
