摘  要:目的通过建立动物的近视眼模型,观察近视眼发展期和恢复期屈光状态的改变以及基质金属蛋白酶(MMP-2)的含量变化,探讨小鸡近视眼模型巩膜重塑过程中的生物化学变化机制。设计随机对照实验性研究。研究对象32只2天龄SPF美国进口白种莱航鸡。方法将32只鸡随机分为A、B、C、D四组,每组8只,A、B组为实验组,用透明眼罩遮盖小鸡的右眼进行单眼形觉视觉剥夺,左眼为对照;C、D组为正常对照。21天后去遮盖,检测双眼屈光状态和眼轴,处死A组和C组并分离7mm直径的后巩膜组织;继续饲养B组和D组至去遮盖后21天,检测双眼屈光状态和眼轴后处死并分离7mm直径的后巩膜组织。用酶活性分析法测定所有动物巩膜中MMP-2的含量,并将实验组和对照组的数据进行统计学分析。主要指标遮盖后及去遮盖后小鸡的屈光度、眼轴,巩膜中MMP-2的含量。结果遮盖21天A、B组左右眼屈光度比较,差异有统计学意义(P= 0.000),平均相差(-28.38±6.72)D;A组左右眼眼轴相比差异有统计学意义(P=0.000),平均相差(1.47±0.37)mm;遮盖眼后极部巩膜中的MMP-2含量显著性升高,A组遮盖眼与对侧无遮盖眼间比较差异有统计学意义,平均增加(76.00±15.20)%。去遮盖21天B组左右眼屈光度比较差异无统计学意义,平均差值(0.19±0.92)D;B组左右眼眼轴比较差异无统计学意义,平均差值(0.15±0.50)mm;去遮盖眼后极部巩膜中的MMP-2含量明显下降,与对照组之间差异无统计学意义(P=0.47)。结论形觉剥夺性近视眼的小鸡模型中,近视眼的形成与后巩膜中的MMP-2水平有关,而且MMP-2的含量在近视眼发展期和恢复期存在一定的变化规律。(眼科,2006,15:334-337)Objective To study the biochemical change in the sclera of FDM (form-deprived myopia) chicks during the process of sclera remodeling, and assess the role of this metalloproteinase (MMP-2) in ocular elongation associated with experimental myopia and the dynamic change of it. Design Randomized, controlled experimental study. Participant 32 experimental white Leghorn chicks. Method 32 2-day-old white Leghorn chicks were divided into four groups randomly. FDM was induced by applying translucent plastic goggles attached on the right eyes-Group A and B. Untreated fellow eyes served as paired controls, and normal controls too-Group C and D were set. The goggles were kept in place for 21 days, and all the eyes were checked up for refractive condition. Then killed the chicks of Group A and C, and the sclera punches (7mm in diameter) were excised from the posterior poles of control and FDM eyes with surgery trephine after measuring the axis of the eyes. Half of the treated chicks (Group B) were raised for 21 days more. All the eyes were checked up for refractive condition and axial length. Sclera punches (7 mm in diameter) were also excised from the posterior poles. The amount of MMP-2 was measured with activity assay and the data were analyzed with SPSS 11.0. Main Outcome Measure The refraction results, axis of the eyes, and the contents of MMP-2 in the posterior sclera. Result After deprivation for 21 days, significant differences were detected in refractions between covered eyes and untreated eyes (P=0.000), the average difference was -28.38± 6.72D. Significant differences were also detected in eye elongations(P=0.000), the average difference was 1.47±0.37mm. MMP-2 activity in deprived posterior sclera was 76.00±15.20% higher than normal controls when treated with APMA. After recovery from FDM no significant differences were found in refraetions and eye elongations between treated eye and normal control. And the MMP-2 also decreased to normal level compared with controls (P=0.47). Conc
