抑癌基因PTEN和p27^(kip1)在涎腺腺样囊性癌中的表达及意义

摘　　要：目的研究抑癌基因PTEN、p27k ip1在涎腺腺样囊性癌(salivary adenoid cystic carc inom a,SACC)中的表达及其相关性,探讨PTEN、p27k ip1表达在涎腺腺样囊性癌的诊断和判断预后中的价值.方法应用免疫组化SP染色法检测PTEN、p27k ip1在涎腺腺样囊性癌、涎腺多形性腺瘤中的表达.结果PTEN、p27k ip1在涎腺腺样囊性癌中存在明显表达缺失或表达降低.在SACC中PTEN的阳性表达率和表达强度与病理学分型有关,与患者的年龄和临床分期无关.p27k ip1表达缺失与SACC患者的年龄、临床分期和病理学分型无关.SACC中PTEN与p27k ip1表达呈负相关性.结论PTEN和p27k ip1在涎腺腺样囊性癌中明显存在表达缺失或表达降低,PETN的表达异常与涎腺腺样囊性癌的病理学分型有关,恶性程度越高PTEN表达越低.p27k ip1表达强度可用于辅助涎腺腺样囊性癌与涎腺良性肿瘤的鉴别诊断.SACC中PTEN与p27k ip1表达呈负相关性.[Abstract] Objective To determined the potential role of PTEN and p27^kip1 expression in the diagnosis and prognosis of adenoid cystic carcinoma. Method Expression of PTEN and p27^kip1 were detected by immunohistochemical staining SP method. Results Loss of PTEN and p27^kip1 expression were significant in adenoid cystic carcinoma. In adenoid cystic carcinoma PTEN - staining status were associated with pathematology classing but not associated with patient＇s age and TNM classification. P27^kip1 -staining status was not associated with patient＇s age, TNM classification and pathematology classing, Loss of PTEN expression was not correlated with that of p27^kip1 expression in adenoid cystic carcinoma. Conclusions Loss of PTEN and p27^kip1 expression were significant in adenoid cystic carcinoma. In adenoid cystic carcinoma, PTEN- staining status was associated with pathematology classing, the higher malignancy degree, the lower expression of PTEN. The p27^kip1 may be helpful to differentiate SACC from benign salivary tumors. There was a negative correlation between the expression of PTEN and p27^kip1.
