摘 要:目的优化急性肝衰竭模型的构建方法,探讨细胞移植的最佳途径。方法以D-氨基半乳糖胺(D-gal)作为肝脏毒剂,构建雌性SD大鼠急性肝衰竭模型。以病理形态学和多项血生化指标综合评估肝功能损伤的状态,确定模型构建的最优方案。以此为基础,通过脾内和腹股沟静脉两种途径植入雄性SD大鼠的骨髓间质干细胞(BMSCs)。以sry基因为分子标志,采用PCR技术检测植入细胞在肝脏内的存活状态,并对两种不同的植入途径综合评估。结果(1)不同剂量的D-gal可导致不同程度的肝损伤,而1.4g/kg剂量较好地模拟了急性肝衰竭的病理生理改变且稳定性较好。(2)移植后7~30天在两组细胞移植组大鼠肝脏内均能检测到sry基因阳性细胞的存在,而相应对照组未能检测到sry基因阳性细胞的存在。结论(1)D-gal 1.4g/kg腹腔注射可以很好构建SD大鼠急性肝衰竭模型。(2)骨髓间质细胞经腹股沟静脉移植和脾内移植均可在受体肝脏内存活至少1个月。经腹股沟静脉进行移植是治疗大鼠急性肝衰竭的一种较好的途径。Objective To optimize the way of establishing a rat model of acute hepatic failure, and preliminarily study the way of cell transplantation. Methods A female rat model of liver injury induced by D - galactosamine ( D - gal ) was established. The degree of liver injury was evaluated according to pathology morphology and the levels of biochemistry to find the best project. Based on it, male rats bone marrow mesenchymal stem cells(BMSCs) were transplanted through spleen and groin vein. With the molecule symbol of sry gene , the live ability of planted cells in the liver of female recipients was explored by PCR , and the different transplanted ways were evaluated. Results (1)Different extent hver injury can be induced by different dosages of D - gal, and the pathophysiology of acute hepatic failure can be copied very well at the dose of 1.4g/kg, and the model is very steady. (2) BMSCs transplantation in both experimental groups can lower the death rate of acute hepatic failure rats .Shown by PCR analysis of the liver DNA, the sry gene was positive from 7 days to 30 days in the liver female recipients which had been transplanted with BMSCs from male rats, however was negative in the control groups. Conclusion ( 1 ) Acute hepatic failure of rats can be induced with D - gal at the dose of 1.4g/kg very well. (2)BMSCs can survive in the liver of recipients both intraspleen and intra - groin - venous transplantation, but the death rate is lower in the group of intra- groin- venous transplantation than in the group of intraspleen transplantation, and the operation in the group of intra- groin- venous transplantation is very easy, so intra- groin - venous transplantation is a better way to treat acute hepatic failure of rat.
